  1. 1. A Chemical Approach Toward the Therapeutic Optimization of Cytotoxic Bioconjugates

    Author : Rafael Hartmann; Luke R. Odell; Fredrik Lehmann; Mats Larhed; Dario Neri; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Chemotherapy; antibody-drug conjugates; organic synthesis; bioorganic chemistry; synthetic method development; analytical method development; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi; Medicinal Chemistry; Läkemedelskemi; Chemistry with specialization in Organic Chemistry; Kemi med inriktning mot organisk kemi;

    Abstract : The delivery of cytotoxicity to specific cells was recognized to be the key to the effective, chemical treatment of infectious and neoplastic diseases well over a century ago. In the context of oncology, the primary therapeutic challenge is the distinction between healthy and pathological cells despite their pronounced pheno- and genotypic similarity. READ MORE

  2. 2. Peptide Conjugates as Useful Molecular Tools

    Author : Adam T. Ślósarczyk; Lars Baltzer; Knud J. Jensen; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; peptide conjugates; designed peptides; polypeptides; molecular recognition; linker; linking technology; bioconjugation; conjugation; conjugates; Kemi; Chemistry; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi;

    Abstract : The conjugation of a small organic molecule to synthetic polypeptides from a designed set has been shown to give rise to binders with high affinity and selectivity for the phosphorylated model proteins α-casein and β-casein but not for ovoalbumin. The small organic molecule that was used for this purpose is comprised of two di-(2-picolyl)amine groups assembled on a dimethylphenyl scaffold, and is capable of complexing two Zn2+ ions to form chelates that bind the phosphate ion. 4. 4. Conformationally Constrained Oligonucleotides for RNA Targeting

    Author : Qing Li; Jyoti Chattopadhyaya; Li-He Zhang; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; conformationally constrained nucleoside; antisense oligonucleotide; RNA affinity; nuclease stability; RNase H; RNA degradation; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi;

    Abstract : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. READ MORE
