  1. 1. Protein production and purification in structural genomics

    Author : Martin Hammarström; Per-Åke Nygren; Torleif Härd; Helena Berglund; Herman van Tilbeurgh; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Recombinational cloning; Escherichia coli; gene expression; fusion proteins; solubility; circular dichroism; nuclear magnetic resonance; Structural biochemistry; Strukturbiokemi;

    Abstract : The number of gene products available for structural and functional study is increasing at an unprecedented rate as a result of the successful whole genome sequencing projects. Systematic structure determination of proteins on a genomic scale, called structural genomics, can significantly contribute to the field of protein science and to functional annotation of newly identified genes. 2. 2. Selection of antigens for antibody-based proteomics

    Author : Lisa Berglund; Mathias Uhlén; Sören Brunak; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; epitope; antigen; antibody; affinity; protein; proteome; proteomics; bioinformatics; prediction; primer design; sequence similarity; Bioengineering; Bioteknik;

    Abstract : The human genome is predicted to contain ~20,500 protein-coding genes. The encoded proteins are the key players in the body, but the functions and localizations of most proteins are still unknown. READ MORE

  3. 3. Bacterial display systems for engineering of affinity proteins

    Author : Filippa Fleetwood; John Löfblom; Harald Kolmar; KTH; []
    Keywords : Combinatorial protein engineering; staphylococcal display; Affibody; biparatopic; VEGFR2; nanobody; E. coli display; autotransporter; Bioteknologi; Biotechnology;

    Abstract : Directed evolution is a powerful method for engineering of specific affinity proteins such as antibodies and alternative scaffold proteins. For selections from combinatorial protein libraries, robust and high-throughput selection platforms are needed. READ MORE

  4. 4. Crustacean immunity : Characterization of some crayfish blood proteins

    Author : Pia Gambäck Keyser; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Plant physiology; Växtfysiologi; Plant physiology; Växtfysiologi; fysiologisk mykologi; Physiological Mycology;

    Abstract : The cDNA sequence and some properties of a previously purified #x03B2;-1,3-glucan binding protein are reported here. The putative amino acid sequence (predicted molecular mass: 152 kDa) of the protein did not show any significant similarity to any known protein, but an RGD motif and a region with slight similarity to bacterial glucanase was found. 5. 5. Fighting Tuberculosis – : Structural Studies of Three Mycobacterial Proteins

    Author : Alina Castell; Torsten Unge; Manfred Weiss; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Mycobacterium tuberculosis; Rv0216; Rv0130; Mycobacterium smegmatis; branched chain aminotransferase; X-ray crystallography; fatty acid metabolism; Structural biology; Strukturbiologi;

    Abstract : This thesis presents the cloning, purification, crystallization, and structural studies of two unknown proteins from Mycobacterium tuberculosis, and of an aminotransferase from Mycobacterium smegmatis. Structural knowledge of these proteins is of highest interest for structure-based drug design, which is one of the approaches that can be used in order to fight tuberculosis (TB). READ MORE
