  1. 1. Detection and Sequencing of Amplified Single Molecules

    Author : Rongqin Ke; Mats Nilsson; Ulf Landegren; Radoje Drmanac; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; padlock probes; proximity ligation assay; rolling circle amplification; circle-to-circle amplification; single molecule detection; amplified single molecules; sequencing; in situ; single cell; Molecular Biotechnology; Molekylär bioteknik;

    Abstract : Improved analytical methods provide new opportunities for both biological research and medical applications. This thesis describes several novel molecular techniques for nucleic acid and protein analysis based on detection or sequencing of amplified single molecules (ASMs). READ MORE

  2. 2. Novel planar and particle-based microarrays for point-of-care diagnostics

    Author : Gustav Svedberg; Helene Svahn Andersson; Jesper Gantelius; Fridtjof Lund-Johansen; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Planar microarrays; Particle-based microarrays; Point-of-care diagnostics; Colorimetric detection; Signal enhancement; Isothermal DNA amplification; Solid-phase DNA amplification; Biotechnology; Bioteknologi;

    Abstract : Point-of-care assays are easy-to-use, portable and inexpensive tests that canbe used to aid diagnostics by measuring levels of disease-specific moleculesin settings where access to advanced laboratory equipment and trainedpersonnel are limited, such as at the patient's bedside or in low resourceparts of developing countries. In order to achieve high multiplexingcapacities, such assays can be based on planar microarrays consisting ofspots immobilized on a flat surface or on particle-based microarrays basedon populations of encoded particles. 3. 3. Novel methods for improving rapid paper-based protein assays with gold nanoparticle detection

    Author : Lara Lama; Helene Andersson Svahn; Jesper Gantelius; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Gold nanoparticles; ultrasound energy; signal enhancement; diagnostics; microarrays; paper-based assays; COMSOL Multiphysics simulations; Bioteknologi; Biotechnology;

    Abstract : This thesis describes methods for improving sensitivity in rapid singleplex and multiplex microarray assays. The assays utilize the optical characteristics of colloidal gold nanoparticles for the colorimetric detection of proteins.Multiplexed detection in sandwich immunoassays is limited by cross-reactivity between different detection antibodies. 4. 4. Novel microfluidic based sample preparation methods for rapid separation and detection of viable bacteria from blood for sepsis diagnostics

    Author : Sharath Narayana Iyengar; Aman Russom; Jonas Tegenfeldt; KTH; []
    Keywords : microfluidics; Bioteknologi; Biotechnology;

    Abstract : Sepsis is a serious medical condition characterized by a whole-body inflammatory response caused by bloodstream infection. The final stage of sepsis can lead to septic shock, multiple organ failure, and death. In early sepsis, the concentration of bacteria in the bloodstream is typically low, making diagnosis challenging. READ MORE

  5. 5. Molecular diagnostics of infectious diseases

    Author : Malin Nygren; KTH; []
    Keywords : ;

    Abstract : In DNA-based diagnostics, the polymerase chain reaction(PCR) is the most widely used DNA amplification method. Toenable both sensitive and specific detection of agents causinginfectious diseases, the PCR needs to becombined with methodsto prepare the clinical sample containing the genetic materialof the pathogen.
