  1. 1. Towards unbiased 3D reconstruction : in single-particle cryo-electron microscopy

    Author : Dominika Elmlund; Hans Hebert; Sarah Butcher; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Bioengineering; Bioteknik;

    Abstract : Cryo-electron microscopy of freestanding molecules (single-particles) plays a pivotal role in the difficult and pressing challenge of determining the structures of large macromolecular complexes. Molecular volumes are generated by aligning large sets of randomly oriented two-dimensional (2D) projection images in three dimensions (3D) before reconstruction is performed using tomographic techniques. 2. 2. Revealing the architecture and composition of the sperm flagellum tip

    Author : Davide Zabeo; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Cell biology; Structural biology; Microtubules; Electron microscopy; Cryo-electron microscopy; Cryo-electron tomography;

    Abstract : The eukaryotic flagellum is a membrane-bound protruding organelle with a cytoskeleton of microtubules. Flagella are found in unicellular as well as multicellular organisms, performing a variety of functions. READ MORE

  3. 3. Cryo-electron tomography of individual protein molecules

    Author : Sara Sandin; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Cryo-Electron Microscopy; Cryo-Electron Tomography; Protein Conformation; Antibodies; Immunoglobulin G; Glucocorticoid Receptor; Hepatocyte Growth Factor Scatter Factor; Receptor Tyrosine Kinase MET;

    Abstract : Averaging methods of determining structure, such as X-ray diffraction, do not preserve information about the flexibility of molecules. Cryo-electron tomography allows us to reconstruct individual hydrated objects. READ MORE

  4. 4. Single-particle cryo-electron microscopy of macromolecular assemblies

    Author : Kimberley Cheng; Hans Hebert; Roland Beckmann; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Bioengineering; Bioteknik;

    Abstract : In this thesis, single-particle cryo-electron microscopy (cryo-EM) was used to study the structure of three macromolecular assemblies: the two hemocyanin isoforms from Rapana thomasiana, the Pyrococcus furiosus chaperonin, and the ribosome from Escherichia coli. Hemocyanins are large respiratory proteins in arthropods and molluscs. READ MORE

  5. 5. Contributions to 3D Image Analysis using Discrete Methods and Fuzzy Techniques : With Focus on Images from Cryo-Electron Tomography

    Author : Magnus Gedda; Ewert Bengtsson; Stina Svensson; Carolina Wählby; Punam Saha; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; digital image analysis; 3D; fuzzy; algorithms; grey-weighted distance; region growing; electron tomography; tracing; fluorescence microscopy; Datoriserad bildanalys; Computerized Image Analysis;

    Abstract : With the emergence of new imaging techniques, researchers are always eager to push the boundaries by examining objects either smaller or further away than what was previously possible. The development of image analysis techniques has greatly helped to introduce objectivity and coherence in measurements and decision making. READ MORE
