  1. 1. Methods for Preclinical Evaluation of Cytotoxic Drugs : With Special Reference to the Cyanoguanidine CHS 828 and Hollow Fiber Method

    Author : Saadia Bashir Hassan; Elin Lindhagen; Rolf Larsson; Mats O Karlsson; Mustapha Hassan; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacology; CHS 828; in vitro; in vivo; hollow fiber; models; NF-kB; Farmakologi; Pharmacological research; Farmakologisk forskning;

    Abstract : The novel cyanoguanidine CHS 828 has shown promising antitumor activity in many in vitro and in vivo studies. The long-term 14 days in vitro hollow fiber cultures, where tumor cells from different tumor cell lines were cultured inside semipermeable fibers, were more resistant to CHS 828 and other cytotoxic drugs than the shorter-term 3 days cultures. 2. 2. Studies of Cytotoxic Compounds of Natural Origin and their Mechanisms of Action

    Author : Jenny Felth; Lars Bohlin; Joachim Gullbo; Ulf Göransson; Thomas Efferth; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cytotoxic; cardiac glycoside; gambogic acid; cancer; mechanism of action; Pharmacognosy; Farmakognosi; Pharmacognosy; Farmakognosi;

    Abstract : Cancer incidence is increasing and novel anticancer drugs with new mechanisms of action are essential for future chemotherapeutic treatment. Natural products have historically played an important role in the development of anti-cancer drugs and have potential to do so also in the future. READ MORE

  3. 3. Studies on Cytotoxic and Neutrophil Challenging Polypeptides and Cardiac Glycosides of Plant Origin

    Author : Senia Johansson; Tore Mellstrand; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical chemistry; cytotoxic; neutrophil; polypeptides; Phoradendron tomentosum; Digitalis purpurea; plant; mistletoe; MS MS; Edman degradation; breast cancer; macrocyclic peptides; multidrug resistance protein; primary structures; phoratoxin; isolation; digitoxin; cardiac lycoside; structure-activity; Farmaceutisk kemi; Pharmaceutical chemistry; Farmaceutisk kemi; Pharmacognosy; farmakognosi;

    Abstract : This thesis examines the isolation and characterisation (biological and chemical) of polypeptides from plants. A fractionation protocol was developed and applied on 100 plant materials with the aim of isolating highly purified polypeptide fractions from small amounts of plant materials. READ MORE

  4. 4. Exploring Cancer Drugs In Vitro and In Vivo : With Special Reference to Chemosensitivity Testing and Early Clinical Development

    Author : Anne von Heideman; Peter Nygren; Bengt Tholander; Rolf Larsson; Seija Grénman; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cytotoxic drug; in vitro assay; drug development; phase I clinical trial; Oncology; Onkologi; Oncology; Onkologi;

    Abstract : The aims of this thesis were to investigate the utility of in vitro drug sensitivity testing to optimize the use of cancer chemotherapy and to assess the properties of a new cancer drug in a phase I clinical trial. Tumour cells from patients were analysed with the short-term Fluorometric Microculture Cytotoxicity Assay (FMCA). READ MORE

  5. 5. Development and application of new methods for anticancer drug discovery and early evaluation

    Author : Sumeer Dhar; Uppsala universitet; []
    Keywords : Medical sciences; Drug development; tumor cell lines; cytotoxic drugs; neural network; MEDICIN OCH VÅRD; MEDICINE; MEDICIN; klinisk farmakologi; Clinical Pharmacology;

    Abstract : A non-clonogenic assay using the redox dye Alamar Blue and primary human tumor cells (PHTC) from patients and human tumor cell lines (HTCL) was established for rapid screening and evaluation of anticancer drugs. PHTC cultures from chronic lymphocytic leukemia (CLL) and ovarian carcinoma (Ovca) were compared with renal carcinoma (ACHN) and lymphocytic (CCRF-CEM) HTLCs in response to 63 toxic or non-toxic compounds.
