  1. 1. Quantum Chemical Studies of Chemotherapeutic Drug Cisplatin : Activation and Binding to DNA

    Author : Johan Raber; David van der Spoel; Paolo Carloni; Uppsala universitet; []
    Keywords : Quantum chemistry; cisplatin; quantum chemistry; DNA; density functional theory; activation; JM118; substitution reaction; Platinum; guanine; adenine; cytostatic drug; aquation; anation; activation; Kvantkemi;

    Abstract : The serendipitous discovery of the potent cytotoxic properties of cisplatin brought about a revolution in the treatment of certain types of cancer, but almost fifty years later, there still remain unknown areas in the chemistry of cisplatin. There are questions regarding which form of the drug reaches its DNA target, or why certain DNA sequences are more preferred than others for reaction with cisplatin. READ MORE

  2. 2. Biomarkers of internal exposure/dose : Methods to quantify adducts to protein and DNA by LC/MS studied with benzo[a]pyrene and isocyanates

    Author : Emelie Westberg; Margareta Törnqvist; Kimmo Peltonen; Stockholms universitet; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; biomarker; PAH; serum albumin; adduct; DNA; isocyanate; Environmental Chemistry; miljökemi;

    Abstract : This thesis focuses on methods for quantification by liquid chromatography/mass spectrometry (LC/MS) of specific biomarkers for internal dose of chemicals which induce toxicity through their electrophilic reactivity. In vivo such compounds are short-lived, and could feasibly be measured as their reaction products (adducts) with biomacromolecules. READ MORE

  3. 3. Nucleic Acids as Drug Targets -Interactions of Platinum Complexes, Polyamines, and siRNA with DNA and RNA Models

    Author : Margareta Hägerlöf; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Kinetics; Kemi; Chemistry; AU-rich region; siRNA; Circular Dichroism; Polyamine; Modified Nucleoside; HPLC; Thermodynamic Stability; cisplatin; Hairpin; DNA and RNA conformation; RNA; DNA; Anticancer Drug; Platinum Complexes;

    Abstract : Drugs targeting nucleic acids were studied in DNA and RNA model systems. The effects were investigated with respect to structural and thermodynamic influence, kinetics, and biological function. The drugs employed in this thesis were anticancer active platinum(II) complexes, polyamine derivatives, and siRNA. READ MORE

  5. 5. Studies on DNA adducts and carcinogenesis with 2-nitrofluorene as a model substance

    Author : Xian-Shu Cui; Karolinska Institutet; Karolinska Institutet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; 2-nitrofluorene; carcinogens; DNA adducts; preneoplastic lesion; tumor;

    Abstract : Studies on DNA Adducts and Carcinogenesis with 2-Nitrofluorene as a Model Substance Xian-Shu Cui Center for Nutrition and Toxicology Department of Biosciences at Novum, Karolinska institute Novum, S-141 57 Huddinge, Sweden A number of carcinogens or their electrophilic intermediates are able to bind covalently to DNA, forming DNA adducts. DNA adducts may cause gene mutations if they are misrepaired or remain unrepaired before DNA replication occurs. READ MORE
