  1. 1. Oxidative damage and the DNA glycosylase MutYH

    Author : Kristina Jansson; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Oxidative damage; 8-oxo-7; 8-dihydro-2′-deoxyguanosine; Base excision repair; DNA glycosylase; Mismatch repair; DNA damage checkpoint; 9-1-1 complex; Glutathione-S-transferases; Colon cancer.;

    Abstract : The DNA glycosylase MutYH is highly conserved throughout evolution, and homologs are found in most eukaryotes and prokaryotes examined. MutYH functions as a base excision repair DNA glycosylase that excises adenines misincorporated opposite 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxodG), one of the most stable products of oxidative DNA damage. 2. 2. Targeting DNA repair pathways for cancer therapy

    Author : Saeed Eshtad; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Accumulation of genomic mutations is the consequence of failure in DNA repair as well as increased exposure to endogenous/environmental mutagens. DNA repair pathways safeguard the human genome from such mutagens, and thereby suppress the multi-step process of carcinogenesis. READ MORE

  3. 3. Computational chemistry studies of UV induced processes in human skin

    Author : Jonas Danielsson; Aatto Laaksonen; Villy Sundström; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Photochemistry; Theoretical chemistry; Density functional theory; UV effects; TD-DFT; enzyme catalysis; DNA damage; Molecular dynamics; Physical chemistry; Fysikalisk kemi;

    Abstract : This thesis presents and uses the techniques of computational chemistry to explore two different processes induced in human skin by ultraviolet light. The first is the transformation of urocanic acid into a immunosuppressing agent, and the other is the enzymatic action of the 8-oxoguanine glycosylase enzyme. READ MORE

  4. 4. The DNA repair enzymes MTH1 and OGG1 as targets to treat inflammation

    Author : Stella Karsten; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Chronic and acute inflammatory diseases, such as multiple sclerosis (MS), psoriasis and sepsis, account for vast disability and morbidity in the world. Several new immunomodulating treatment alternatives have been developed over the past decades, but there is still an urgent need for new options. READ MORE

  5. 5. Modulating the activity of OGG1 using small molecules to target the oxidative DNA damage response in cancer and inflammation

    Author : Bishoy M F Hanna; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The production of reactive oxygen species (ROS) is increased in several pathological conditions including cancer and inflammation. Multiple lines of evidence suggest that ROS are involved in signaling events that promote tumorigenesis and inflammatory responses. READ MORE
