1. Nucleic Acids as Drug Targets -Interactions of Platinum Complexes, Polyamines, and siRNA with DNA and RNA Models
Abstract : Drugs targeting nucleic acids were studied in DNA and RNA model systems. The effects were investigated with respect to structural and thermodynamic influence, kinetics, and biological function. The drugs employed in this thesis were anticancer active platinum(II) complexes, polyamine derivatives, and siRNA. READ MORE

2. Interactions of Anticancer Active Platinum(II) Complexes with DNA
Abstract : The kinetics and mechanism for interactions of the anticancer active platinum(II) complexes cis-[PtCl2(NH3)2)], cis-[PtCl2(NH3)(c-C6H11NH2)], and trans-[PtCl2-(NH3)(quinoline)] with DNA have been studied using a combination of HPLC, circular dichroism (CD), UV/vis spectroscopy, and differential scanning calorimetry (DSC). Reactions of the monoaqua forms (charge = +1) of these complexes with single-stranded DNA oligonucleotides, typically between 13-17 bases long, were found to depend on i) the composition of the reaction medium, ii) the exact nature and location of the platination target, and iii) on the DNA structure. 3. Activation of Herpes simplex virus type 1 origin of DNA replication
Abstract : Herpes simplex virus type 1 has been used to study the molecular mechanism of DNA replication. Initiation of DNA replication takes place at specific regions called origins of replication. The HSV-1 genome contains three origins of replication, one copy of oriL and two copies of oriS. READ MORE

4. Mechanisms and DNA Specificity in Site-specific Recombination of Integron Cassettes
Abstract : Bacterial resistance to antibiotics has become a serious problem. This is due to the remarkable ability of bacteria to respond and rapidly adapt to environmental changes. Integrons are elements with the capacity for gene capture by an integron-encoded site-specific recombinase called IntI. READ MORE

5. Herpes Simplex Virus 1 DNA replication and its role in recombination and transcription
Abstract : Herpes simplex virus 1 (HSV-1) is one of nine different herpesvirus infecting man. They are all capable of establishing a life-long latent state following the primary infection. HSV-1 as well as other herpesviruses may reactivate from the latent state and give rise to a productive infection with clinical symptoms or asymptomatic shedding. READ MORE
