  1. 1. Mismatch repair deficiency in colorectal cancer : prognosis and prediction for basic treatment strategies

    Author : Ioannis Gkekas; Jan Novotny; Karin Strigård; Richard Palmqvist; Ulf Gunnarsson; Christian Sturesson; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; colorectal cancer; defective mismatch repair; prognosis and prediction; Oncology; onkologi;

    Abstract : Colorectal cancer (CRC) remains a significant healthcare problem worldwide, being the third most common cancer and the fourth most frequent cause of cancer death. Environmental and dietary factors such as alcohol abuse, cigarette smoking, and genetic predisposition seem to constitute the main aetiologies. READ MORE

  2. 2. Oxidative damage and the DNA glycosylase MutYH

    Author : Kristina Jansson; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Oxidative damage; 8-oxo-7; 8-dihydro-2′-deoxyguanosine; Base excision repair; DNA glycosylase; Mismatch repair; DNA damage checkpoint; 9-1-1 complex; Glutathione-S-transferases; Colon cancer.;

    Abstract : The DNA glycosylase MutYH is highly conserved throughout evolution, and homologs are found in most eukaryotes and prokaryotes examined. MutYH functions as a base excision repair DNA glycosylase that excises adenines misincorporated opposite 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxodG), one of the most stable products of oxidative DNA damage. 3. 3. DNA precursor asymmetries, Mismatch Repair and their effect on mutation specificity

    Author : Robert Buckland; Andrei Chabes; Juan Méndez; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; DNA Replication Fidelity; Mutations; dNTP pools; Mismatch Repair; Checkpoint; Ribonucleotide Reductase; Msh2; medicinsk biokemi; Medical Biochemistry; molekylärbiologi; Molecular Biology;

    Abstract : In order to build any structure, a good supply of materials, accurate workers and quality control are needed. This is even the case when constructing DNA, the so-called “Code of Life.” For a species to continue to exist, this DNA code must be copied with incredibly high accuracy when each and every cell replicates. 4. 4. Mismatch Repair Deficient Cancer Diagnostic Aspects in Colorectal Cancer and the Role of Urological Cancer in Lynch Syndrome

    Author : Patrick Joost; Bröstcancer-genetik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Mismatch repair; Lynch syndrome; immunohistochemistry; microsatellite instability; heterogeneity; cumulative incidence; colorectal cancer; urothelial cancer; prostate cancer; renal cell cancer;

    Abstract : Mismatch repair (MMR) deficiency is carcinogenic and can either have somatic/sporadic causes (i.e. epigenetic silencing or somatic inactivation) or hereditary causes (Lynch syndrome due to a germline mutation in one of the MMR genes ¬- MLH1, MSH2, MSH6, PMS2). READ MORE

  5. 5. O6-methylguanine-DNA-methyltransferase and DNA mismatch repair in relation to drug resistance in malignant melanoma

    Author : Shuhua Ma; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Melanoma; chemotherapy; dacarbazine; drug resistance; O6-methylguanineDNAmethyltransferase; DNA mismatch repair;

    Abstract : Although the survival rate of patients with malignant melanoma has improved, treatment of the disease poses large problems since disseminated melanoma frequently shows primary resistance to chemotherapy. Resistance to chemotherapy is thus a major clinical problem and a cause of failure in the treatment of metastatic malignant melanoma. READ MORE
