  1. 1. The consequences of DNA lesions for mitochondrial DNA maintenance

    Author : Josefin M. E. Forslund; Sjoerd Wanrooij; Nasim Sabouri; Erik Johansson; Claes Gustafsson; Umeå universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Mitochondria; mitochondrial DNA; DNA replication; DNA lesions; DNA polymerase γ; ribonucleotide incorporation; medicinsk biokemi; Medical Biochemistry;

    Abstract : Eukaryotic cells have their own energy-producing organelles called mitochondria. The energy is stored in the adenosine triphosphate (ATP) molecule and is produced via the oxidative phosphorylation process inside the mitochondria. Thirteen of the essential proteins required for this process are encoded on the mitochondrial DNA (mtDNA). READ MORE

  2. 2. Ribonucleotides in DNA - Application in genome-wide DNA polymerase tracking and physiological role in eukaryotes

    Author : Katrin Kreisel; Göteborgs universitet; Göteborgs universitet; Gothenburg University; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Ribonucleotides; DNA instability; DNA polymerase eta; nuclear DNA; mitochondrial DNA;

    Abstract : The genetic code in the eukaryotic cell is stored in the form of DNA, which is more resistant to hydrolysis than RNA. Replication fidelity and DNA repair mechanisms are in place to ensure genomic integrity to preserve the information encoded. READ MORE

  3. 3. Structure of eukaryotic DNA polymerase epsilon and lesion bypass capability

    Author : Nasim Sabouri; Erik Johansson; Lars Thelander; Ola Hammarsten; Umeå universitet; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; NATURVETENSKAP; NATURAL SCIENCES; DNA polymerase epsilon; DNA replication; Okazaki fragment; Translesion synthesis; DNA lesion; dNTP; Biochemistry; Biokemi;

    Abstract : To transfer the information in the genome from mother cell to daughter cell, the DNA replication must be carried out only once and with very high fidelity prior to every cell division. In yeast there are several different DNA polymerases involved in DNA replication and/or DNA repair. READ MORE

  4. 4. Fixation of Functionalized DNA Nanostructures

    Author : Erik Lundberg; Chalmers University of Technology; []
    Keywords : triplex recognition; dna; self-assembly; gel electrophoresis; nanotechnology; cross-linking; spectroscopy; fixation; fret; click chemistry;

    Abstract : This thesis features results from two different studies on a fully addressable non-repetitive network on the nanometre scale, assembled by synthetic DNA molecules. A bio-inspired molecular approach, based on principles of self-assembly, has brought about a paradigm shift in nanoscience, from a top-down to a bottom-up perspective. READ MORE

  5. 5. Role of yeast DNA polymerase epsilon during DNA replication

    Author : Isabelle Isoz; Erik Johansson; Juhani Syväoja; Umeå universitet; []
    Keywords : AGRICULTURAL SCIENCES; LANTBRUKSVETENSKAPER; LANTBRUKSVETENSKAPER; AGRICULTURAL SCIENCES; DNA polymerase epsilon; eukaryotic DNA replication; fidelity; leading strand; Dpb2; Biochemistry and clinical chemistry; Biokemi och klinisk kemi;

    Abstract : Each cell division, the nuclear DNA must be replicated efficiently and with high accuracy to avoid mutations which can have an effect on cell function. There are three replicative DNA polymerases essential for the synthesis of DNA during replication in eukaryotic cells. READ MORE
