  1. 6. Sample preconcentration in open microchannels : Combinations with MALDI and nano-ESI mass spectrometry and computer simulations

    Author : Saara Mikkonen; Åsa Emmer; Leopold Ilag; Jenny Ottosson Takanen; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Preconcentration; open microchannel; microchip; MALDI; electrospray ionization; nanoelectrospray; mass spectrometry; computer simulation;

    Abstract : In this thesis a novel concept for preconcentration of biomolecules in open microchannels is presented. The preconcentration is based on electromigration of charged analytes, and detection is performed with matrix-assisted laser desorption/ionization (MALDI) or nano-electrospray ionization (nESI) mass spectrometry (MS). 2. 7. Method and technique development in peptide and protein mass Spectrometry

    Author : Kristina Håkansson; Uppsala universitet; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Materials science; electrospray ionization; peptide sequencing; Fourier transform ion cyclotron resonance; hydrogen deuterium exchange; protein structure; Materialvetenskap; Materials science; Teknisk materialvetenskap; Molecular Biotechnology; Molekylär bioteknik;

    Abstract : Mechanistic studies and applicability of several methods and techniques for studying peptides and proteins with mass spectrometry are presented. A model for the protein molecular ion formation in explosive matrix-assisted plasma desorption mass spectrometry is proposed, involving a contribution from the chemical energy release during conversion of the explosive, an enhanced protonation through collisions with products from the explosive decay, and electron scavenging by other products. Abstract :  Mass spectrometry (MS) is an important analytical tool. Its most importantadvantages are sensitivity, resolution, and accurate and precise m/zmeasurements. To get the best results from an instrument, the user musthave knowledge of its limitation and requirements. READ MORE

  4. 9. Mass spectrometry of biopolymers based on ion-induced desorption

    Author : Roman Alexandrovich Zubarev; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Radiation sciences; Strålningsvetenskap; Radiation biology; Strålningsbiologi; Ion Physics; jonfysik;

    Abstract : Various aspects of biopolymer molecular characterization by their molecular masses havebeen studied, with emphasis on ion-induced desorption and time-of-flight mass Spectrometry(TOF MS). Limitations of the average-isotopic mass concept, as well as the utility ofaccurate monoisotopic mass measurements for deriving amino-acid and elementalcompositions, are discussed. READ MORE

  5. 10. Development and application of alternative methods for profiling proteins N-glycosylation

    Author : Alessandro Quaranta; Leopold Ilag; Niclas Karlsson; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; N-glycosylation; Glycomics; Glycosylation Biomarkers; Intact Glycoproteins; Glycoform Quantification; Mass Spectrometry; Ion Mobility Spectrometry; MALDI-MS; Paper Spray Ionization; Microfluidics; Magnetic Beads; Immunoaffinity Purification; Nanobodies; Analytical Chemistry; analytisk kemi;

    Abstract : Glycosylation is a post-translational modification (PTM) that exerts profound structural and functional effects on the modified protein. Glycan synthesis and conjugation to proteins are regulated by a myriad of factors, both genetic and environmental, and are also influenced by external stressors. READ MORE
