  2. 2. The genetic background of gestational diabetes mellitus

    Author : Anastasia Papadopoulou; Diabetes och celiaki; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Gestational diabetes mellitus; HLA; TCF7L2; PTPN22; Post partum diabetes; Type 1 Diabetes; Type 2 Diabetes; Islet Cell Autoantibodies;

    Abstract : The aims of this work is to better determine the genetic background of gestational diabetes (GDM) and to examine how specific genes affect the development of diabetes post partum. In the DiPiS (Diabetes Prediction in Skåne) study we typed for HLA-DQB1 alleled, the transcription factor 7-like 2(TCF7L2) rs7903146, rs12255372 and rs7901695 SNPs, the 1858 C>T SNP of the protein tyrosine phosphatase non-receptor 22 (PTPN22) gene and we tested for the presence of islet cell autoantibodies against glutamic acid decarboxylase 65 (GAD65), insulinoma-associated antigen 2 (IA-2) and insulin in women who had been diagnosed with GDM at least once during 2000-2004. Abstract : Pharmacometric modelling is widely used in many aspects related to type 2 diabetes mellitus (T2DM), for instance in the anti-diabetes drug development, and in quantifying the disease progression of T2DM.The aim of this thesis were to improve the design of early phase anti-diabetes drug development studies with the focus on the power to identify mechanism of drug action (MoA), and to characterize and quantify the progression from prediabetes to overt diabetes, both the natural progression and the progression with diet and exercise interventions, using pharmacometrics modelling. 4. 4. Genetic and Metabolic Markers for the Development of Diabetes after Gestational Diabetes Mellitus

    Author : Magnus Ekelund; diabetes och endokrinologi Genomik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Oral glucose tolerance tests; Genetics; Autoimmunity; Insulin sensitivity; Gestational diabetes mellitus; Insulin secretion; HbA1c; Fasting glucose; FTO-gene; Postpartum diabetes;

    Abstract : The aims of this work were to determine the prevalence of postpartum diabetes among women with gestational diabetes mellitus (GDM) in a prospective study 5 years postpartum; to characterize these women according to insulin secretion, insulin sensitivity, autoimmunity and genetics; and to identify possible markers and predictors for the development of manifest diabetes. The subjects were recruited through a regional screening programme in which oral glucose tolerance tests (OGTTs) are routinely offered to all pregnant women. READ MORE

    Abstract : Living with diabetes, as a lifelong illness, is interlaced with learning and to face continual changes. However, the role of time in this learning process is not yet well understood. The overall aim of the thesis was to gain a deepened understanding of learning to live with diabetes for those recently diagnosed and over a three year period. READ MORE
