  1. 1. Neuropeptides and spinal antinociception : studies on galanin, nociceptin and endomorphin

    Author : Stefan Grass; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Endomorphin; flexor reflex; galanin; knock-out mice; neuropathic pain; nociceptin; orphanin FQ transgenic mice;

    Abstract : Nociception and/or the sensation of pain is essential for the survival of the organism. Just as important are the antinociceptive mechanisms that reduce the nociceptive input and perceived pain. Without such mechanisms, it would be impossible to turn ones focus away from even the most minor painful stimulus. READ MORE

  2. 2. Characterization of Substance P (SP) Aminoterminal SP (1-7) Binding in Brain Regions and Spinal Cord of the Male Rat : Studies on the Interaction with Opioid Related Pathways

    Author : Milad Botros; Fred Nyberg; Tsukasa Sakurada; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biological research on drug dependence; Biologisk beroendeforskning;

    Abstract : Binding sites for substance P(1-7), SP(1-7) have been identified and characterized for the first time in crude membrane fraction from rat CNS using tritiated ([3H]) SP(1-7) as tracer. These putative receptors were investigated in relation to their affinity for tachykinins, opioid peptides and sigma receptor ligands. READ MORE

  3. 3. Neuroinflammation and pain

    Author : Linda Block; Göteborgs universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Neuroinflammation; Pain; Persistant pain; Astrocytes; Morphine; Naloxone; Intrathecal administration;

    Abstract : ABSTRACT Background: Persistent pain that remains long after the physiological trigger has been resolved is a disabling condition. A possible mechanism for the transition from acute physiological pain to persistent pain involves low-grade inflammation in the central nervous system, in which inflammatory-activated astrocytes play a significant role. 4. 4. Discovery of Small Peptides and Peptidomimetics Targeting the Substance P 1-7 Binding Site : Focus on Design, Synthesis, Structure-Activity Relationships and Drug-Like Properties

    Author : Rebecca Fransson; Anja Sandström; John S Svendsen; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; substance P 1-7; peptidomimetics; structure-activity relationship; drug-like properties; phenylalanine; imidazole; MAP aryl amides; carbonylation; Pharmaceutical chemistry; Farmaceutisk kemi; Medicinal Chemistry; Läkemedelskemi;

    Abstract : Biologically active peptides are important for many physiological functions in the human body and therefore serve as interesting starting points in drug discovery processes. In this work the neuropeptide substance P 1–7 (SP1–7, H-Arg-Pro-Lys-Pro-Gln-Gln-Phe-OH), which has been demonstrated to reduce neuropathic pain and attenuate opioid withdrawal symptoms in animal models, has been addressed in a medicinal chemistry program with the overall aim of transforming this bioactive peptide into more drug-like compounds. 5. 5. The hippocampal opioid system : role in spatial learning

    Author : Johan Sandin; Karolinska Institutet; Karolinska Institutet; []
    Keywords : opioid; dynorphin; nociceptin; hippocampus; learning; memory; acetylcholine; HPLC; microinjections;

    Abstract : The main aim of the present thesis was to study the role of the endogenous hippocampal opioid system, and in particular, the role of dynorphin B and nociceptin, in spatial learning. The effects of bilateral infusion (using chronic cannulae) of dynorphin B into the dorsal hippocampus (CA3-region), an area containing a high number of dynorphin-containing neurons, were examined in the Morris swim-maze, a spatial task dependent on hippocampal functioning.
