  1. 1. Integration of Aqueous Two-Phase Systems into Recovery Processes for Biomolecules

    Author : Cecilia Kepka; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Proteins; proteins; plasmid DNA; large-scale; protein purification; integration; Expression; aqueous two-phase systems; enzymology; Proteiner; enzymologi;

    Abstract : The main scope of this thesis has been to study and develop new purification methods and also to further develop existing methods for purification to enhance their efficacy. Important parameters in the development of the methods are that they should be suitable for scale-up and possible to be integrated into recovery processes. READ MORE

  2. 2. Production and delivery of recombinant subunit vaccines

    Author : Christin Andersson; KTH; []
    Keywords : Affibody; Affinity chromatography; Affinity purification; DNA immunization; Expression plasmid; Fusion protein; Hydrophobic tag; Iscoms; Lipid tagging; Malaria; Mammalian cell expression; Recombinant immunogen; Respiratory Syncytial Virus; Semliki Fo;

    Abstract : Recombinant strategies are today dominating in thedevelopment of modern subunit vaccines. This thesis describesstrategies for the production and recovery of protein subunitimmunogens, and how genetic design of the expression vectorscan be used to adapt the immunogens for incorporation intoadjuvant systems. 3. 3. CopA and CopT: The Perfect RNA Couple

    Author : Jacoba G. Slagter-Jäger; Gerhart Wagner; Wolfgang Nellen; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Microbiology; antisense RNA; plasmid; U-turn; four-way junction; RNA-RNA interaction; Hfq; Mikrobiologi; Microbiology; Mikrobiologi; mikrobiologi; Microbiology;

    Abstract : Antisense RNAs regulate gene expression in many bacterial systems. The best characterized examples are from prokaryotic accessory elements such as phages, plasmids and transposons. Many of these antisense RNAs have been identified as plasmid copy number regulators where they regulate the replication frequency of the plasmid by negative feedback. READ MORE

  4. 4. Molecular mechanism of plasmid copy number control in Yersinia

    Author : Tifaine Héchard; Helen Wang; Javier Pizarro-Cerda; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Yersinia pseudotuberculosis; plasmid copy number; T3SS; infection; ddPCR; Mikrobiologi; Microbiology;

    Abstract : The ability of pathogenic bacteria to cause disease depends on various virulence mechanisms. The three pathogenic species of Yersinia use a type III secretion system (T3SS) to translocate effector proteins into host cells and disrupt the immune system. This T3SS is encoded on a 70kb, low-copy, virulence plasmid. READ MORE

  5. 5. Temperature-inducible and calcium-regulated proteins encoded by the virulence plasmid of Yersinia

    Author : Ingrid Bölin; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Yersinia; virulence plasmid; gene products; regulation; calcium; temperature; outer membrane proteins;

    Abstract : The pathogenic members of the genus Yersinia, Y. pseudotuberculosis, Y. pestis and Y. enterocolitica are transmitted from animals to man and may give rise to disease with a variety of symptoms. READ MORE
