1. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors
Abstract : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 2. Chimeric gene delivery vectors : Design, synthesis, and mechanisms from transcriptomics analysis
Abstract : Delivery of nucleic acid is a promising approach for genetic diseases/disorders. However, gene therapy using oligonucleotides (ONs) suffers from low transfection efficacy due to negative charges, weak cellular permeability, and enzymatic degradation. READ MORE

3. THE ANDROGEN RECEPTOR GGN POLYMORPHISM. Genetic and functional analyses
Abstract : Male sex development and reproductive function is regulated by androgens acting via the androgen receptor (AR). The AR harbours two polymorphic repeats of CAG and GGN triplets, encoding glutamines and glycines, respectively. READ MORE

4. Genetic re-targeting of adenoviral vectors for gene therapy applications
Abstract : The adenovirus (Ad) is a frequent cause of upper respiratory tract infections, enteritis and conjunctivitis. Since its first isolation in 1953, the study of Ad has contributed greatly to the understanding of intracellular events and has also served as a model system for many viruses. READ MORE

5. Genetic vaccination against acute viral disease
Abstract : This thesis describes the development of recombinant vaccines based on the Semliki Forest virus (SFV) expression system. Immunisation of mice with recombinant virus particles, a layered DNA/RNA plasmid vector, and recombinant self-replicating RNA were carried out and the protective effect of these recombinant vaccines against viral challenge were examined.
