  1. 6. Redesign of Alpha Class Glutathione Transferases to Study Their Catalytic Properties

    Author : Lisa O Nilsson; Ralf Morgenstern; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; glutathione transferase; heterologous expression; heterodimer; hydroxyalkenal; protein redesign; sequential design; dimer-interface interactions; enzyme kinetics; glutathione; dynamic C-terminus; viscosity dependence; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Abstract : A number of active site mutants of human Alpha class glutathione transferase A1-1 (hGST A1-1) were made and characterized to determine the structural determinants for alkenal activity. The choice of mutations was based on primary structure alignments of hGST A1-1 and the Alpha class enzyme with the highest alkenal activity, hGST A4-4, from three different species and crystal structure comparisons between the human enzymes. 2. 7. Multiple Functions of Glutathione Transferases : A Study on Enzymatic Function, Regulatory Role and Distribution in Mouse and Man

    Author : Maryam Edalat; Joseph De Pierre; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Abstract : To cope with various endogenous toxin and xenobiotics nature has equipped the organisms with a proper protection system. Glutathione transferases (GSTs) are important components of the cellular defense against oxidative stress. These proteins appear to be suited for different tasks. READ MORE

  3. 8. Evolutionary Analysis and Posttranslational Chemical Modifications in Protein Redesign : A Study on Mu Class Glutathione Transferases

    Author : Ylva Ivarsson; Bengt Mannervik; Gun Stenberg; Richard N. Armstrong; Uppsala universitet; []
    Keywords : Biochemistry; glutathione transferase; evolutionary analysis; positive selection; epoxide; stereoselectivity; protein modification; Biokemi;

    Abstract : Glutathione transferases (GSTs) constitute a family of multifarious enzymes that conjugate glutathione (GSH) with a wide range of electrophiles. GSTs are grouped into different classes based on protein sequence similarities. Despite high sequence identities between GSTs of the same class they often display different substrate specificites. 4. 9. Catalysis and Site-Specific Modification of Glutathione Transferases Enabled by Rational Design

    Author : Sofia Håkansson Hederos; Lars Baltzer; Kerstin Broo; Linköpings universitet; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Catalysis; Glutathione Transferases; Rational design; Protein design; Site-specific modification; thiolester hydrolysis; Enzyme engineering; Enzymteknik;

    Abstract : This thesis describes the rational design of a novel enzyme, a thiolester hydrolase, derived from human glutathione transferase (GST) A1-1 by the introduction of a single histidine residue. The first section of the thesis describes the design and the determination of the reaction mechanism. READ MORE
