  1. 1. Microsomal glutathione transferase

    Author : Ralf Morgenstern; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : A distinct glutathione transferase which can be activated up to eightfold by sulfhydryl reagents was discovered in rat liver microsomes. This enzyme was subsequently purified to apparent homogeneity in 40 % yield using column cromatography. READ MORE

  2. 2. Microsomal glutathione transferase : Species characteristics, catalysis and topology

    Author : Claes Andersson; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : The membrane-bound detoxication enzyme microsomal glutathione transferase is localized predominantly in the liver of mammalian species, where it is present in high concentrations in the endoplasmic reticulum and the outer mitochondrial membrane. The enzyme conjugates a broad range of hydrophobic xenobiotics bearing reactive electrophilic centers to glutathione, which facilitates deactivation and excretion. READ MORE

  3. 3. Equine glutathione transferase A3-3 : an efficient steroid isomerase

    Author : Helena Lindström; Bengt Mannervik; Ann-Kristin Östlund Farrants; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Equine; glutatione transferase; steroidogenesis; steroid isomerase; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Abstract : Glutathione transferases (GSTs) comprise a superfamily of enzymes prominently involved in detoxication by making toxic electrophiles more polar and therefore more easily excretable. However some GSTs have developed alternative functions. READ MORE

  4. 4. Redesign of Alpha Class Glutathione Transferases to Study Their Catalytic Properties

    Author : Lisa O Nilsson; Ralf Morgenstern; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; glutathione transferase; heterologous expression; heterodimer; hydroxyalkenal; protein redesign; sequential design; dimer-interface interactions; enzyme kinetics; glutathione; dynamic C-terminus; viscosity dependence; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Abstract : A number of active site mutants of human Alpha class glutathione transferase A1-1 (hGST A1-1) were made and characterized to determine the structural determinants for alkenal activity. The choice of mutations was based on primary structure alignments of hGST A1-1 and the Alpha class enzyme with the highest alkenal activity, hGST A4-4, from three different species and crystal structure comparisons between the human enzymes. 5. 5. Directed Enzyme Evolution of Theta Class Glutathione Transferase : Studies of Recombinant Libraries and Enhancement of Activity toward the Anticancer Drug 1,3-bis(2-Chloroethyl)-1-nitrosourea

    Author : Anna-Karin Larsson; Bengt Mannervik; Ian Cotgreave; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; glutathione transferase; directed evolution; 1; 3-bis 2-chloroethyl -1-nitrosourea; alkyltransferase; recombinant libraries; chloroethylnitrosourea; multi-dimensional data analysis; Biokemi; Biochemistry; Biokemi;

    Abstract : Glutathione transferases (GSTs) are detoxication enzymes involved in the cellular protection against a wide range of reactive substances. The role of GSTs is to catalyze the conjugation of glutathione with electrophilic compounds, which generally results in less toxic products. READ MORE
