  1. 1. Fragment Based Drug Discovery with Surface Plasmon Resonance Technology

    Author : Helena Nordström; Helena Danielson; Gregg Siegal; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Fragment based drug discovery; SPR biosensor; matrix metalloproteinase-12; HIV-1 protease; resistance;

    Abstract : Fragment based drug discovery (FBDD) has been applied to two protease drug targets, MMP-12 and HIV-1 protease. The primary screening and characterization of hit fragments were performed with surface plasmon resonance -technology. Further evaluation of the interaction was done by inhibition studies and in one case with X-ray crystallography. READ MORE

  3. 3. Discovery and evaluation of direct acting antivirals against hepatitis C virus

    Author : Eldar Abdurakhmanov; Helena Danielson; David Frick; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Direct acting antivirals; Hepatitis C; NS3-4A protease; NS5B polymerase; structure-based drug discovery; fragment-based drug discovery; surface plasmon resonance; Biokemi; Biochemistry;

    Abstract : Until recently, the standard therapy for hepatitis C treatment has been interferon and ribavirin. Such treatment has only 50% efficacy and is not well tolerated. The emergence of new drugs has increased the treatment efficacy to 90%. READ MORE

  4. 4. Interaction Studies of Secreted Aspartic Proteases (Saps) from Candida albicans : Application for Drug Discovery

    Author : Dan Backman; Helena Danielson; Birgitta Tomkinson; Gunnar Henriksson; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; SPR Biosensor; Secreted aspartic proteases; Candida albicans; interaction kinetics; drug discovery; protease inhibitor; Biokemi; Biochemistry; Biokemi;

    Abstract : This thesis is focused on enzymatic studies of the secreted aspartic proteases (Saps) from Candida albicans as a tool for discovery of anti-candida drugs. C. albicans causes infections in a number of different locations, which differ widely in the protein substrates available and pH. Since C. READ MORE

  5. 5. Structural studies of drug targets and a drug metabolizing enzyme

    Author : Daniela Cederfelt; U. Helena Danielson; Doreen Dobritzsch; Inari Kursula; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Biophysics; Protein structure; X-ray crystallography; Cryogenic electron microscopy; Enzymology; Drug discovery; Pyrimidine degradation; Biokemi; Biochemistry;

    Abstract : The work presented in this thesis describes how structural information about a protein can be acquired, and how it can be used to answer scientific questions about proteins’ function, their dynamic behaviour and their interactions with other proteins or ligands.The catalytic function of the pyrimidine-degrading, drug metabolizing enzyme β-ureidopropionase (βUP) is dependent on the shift between oligomeric states.
