1. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors
Abstract : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 3. Hypoxia inducible factor 1 alpha : dependent and independent regulation of hematopoietic stem cells and leukemia
Abstract : This thesis has studied the role of low oxygen levels, or hypoxia, in hematopoietic stem cells (HSCs) and how, at the molecular level, it regulates stem cell maintenance and protects against oxidative stress induced by reactive oxygen species (ROS). HSCs reside within the bone marrow in specific niches created by a unique vascularized environment, which is suggested to be hypoxic and crucial for HSCs by maintaining a quiescent state of cell cycle and by redirecting metabolism away from the mitochondria to glycolysis. 4. Studies on arachidonic acid metabolism in normal and malignant hematopoietic cells
Abstract : This thesis deals with the metabolism of arachidonic acid via the 5-lipoxygenase- and the 15-lipoxygenase-1 pathways in normal and malignant hematopoietic cells. The first part of the thesis (papers I & II) describes the expression of genes involved in arachidonic acid metabolism and the capacity to generate leukotriene (LT) B4 via the 5-lipoxygenase pathway in blood tumor cells from patients with precursor B-ALL (acute lymphoblastic leukemia) and AML (acute myeloid leukemia).
