  1. 6. Kinetic studies of NS3 and NS5B from Hepatitis C virus : Implications and applications for drug discovery

    Author : Göran Dahl; Helena Danielson; Raffaele De Francesco; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Hepatitis C virus; NS3; NS5B; enzyme kinetics; inhibition; resistance; drug; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : The aim of these studies was to increase our understanding of the non-structural proteins 3 and 5B (NS3 and NS5B) from the hepatitis C virus (HCV), and thereby contribute to the development of new and better drugs against HCV.By studying NS3 with substitutions identified to be associated with resistance to NS3 inhibitors in clinical trials (R155Q, A156T and D168V) it was found that not all inhibitors were affected, indicating that cross-resistance can be avoided. 2. 7. Hepatitis C virus kinetics during antiviral treatment

    Author : Tony Carlsson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Hepatitis C virus RNA; genotype; kinetics; interferon-alpha; ribavirin.;

    Abstract : It is estimated that 170 million individuals have chronic hepatitis C virus (HCV) infection, whereof 20-30% will develop end stage liver disease within 20-30 years. Antiviral therapy can stop this deterioration and clear the infection in 40-80% of the patients depending on treatment and HCV genotype. READ MORE

  3. 8. Towards the elimination of hepatitis C : identifying the infected population, and remaining hepatitis C related risks after successful treatment

    Author : Charlotte Lybeck; Ann-Sofi Duberg; Soo Aleman; Scott Montgomery; Magnus Gottfredsson; Örebro universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; HCV; HCC; epidemiology; MTCT; pregnancy; DAA; OCI;

    Abstract : Chronic Hepatitis C virus (HCV) infection can lead to liver fibrosis and cirrhosis with increased risk of hepatocellular carcinoma (HCC) and liver failure. The World Health Organisation (WHO) has set a goal to eliminate viral hepatitis as a global health threat by 2030. READ MORE

  4. 9. Peptidomimetic Enzyme Inhibitors : Targeting M. tuberculosis Ribonucleotide Reductase and Hepatitis C Virus NS3 Protease

    Author : Johanna Nurbo; Anders Hallberg; Anders Karlén; Anja Sandström; Per Arvidsson; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; enzyme inhibitor; peptidomimetics; structure-activity relationship; tuberculosis; ribonucleotide reductase; hepatitis C virus; NS3 protease; Pharmaceutical chemistry; Farmaceutisk kemi; Medicinal Chemistry; Läkemedelskemi;

    Abstract : This thesis focuses on the design and synthesis of inhibitors targeting Mycobacterium tuberculosis ribonucleotide reductase (RNR) and hepatitis C virus (HCV) NS3 protease; enzymes that have been identified as potential drug targets for the treatment of tuberculosis and hepatitis C, respectively. Small peptides have been recognized as inhibitors of these enzymes. READ MORE

  5. 10. Discovery and evaluation of direct acting antivirals against hepatitis C virus

    Author : Eldar Abdurakhmanov; Helena Danielson; David Frick; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Direct acting antivirals; Hepatitis C; NS3-4A protease; NS5B polymerase; structure-based drug discovery; fragment-based drug discovery; surface plasmon resonance; Biokemi; Biochemistry;

    Abstract : Until recently, the standard therapy for hepatitis C treatment has been interferon and ribavirin. Such treatment has only 50% efficacy and is not well tolerated. The emergence of new drugs has increased the treatment efficacy to 90%. READ MORE
