When a reference genome is missing, highly fragmented or incomplete, a de novo transcriptome assembly is the transcriptome reconstruction approach of choice, since in such situations, a simple alignment (or mapping) would not necessarily give all theinformation concerning splice junctions, isoforms or even the full extent of the gene.     Abstract : The main aim of this thesis was to contribute with detailed information of regional environmental change during the Holocene through studies of speleothems and their growth environments from two caves, Korallgrottan and Labyrintgrottan in northwestern Sweden, and a cellar vault in Uppsala. This was done through studies of stable isotopes and luminescence properties in the speleothems in combination with a detailed monitoring study in Korallgrottan. READ MORE
