  1. 1. Leukotriene C4 synthase : studies on oligomerization and subcellular localization

    Author : Jesper Svartz; Mats Söderström; Sven Hammarström; Colin Funk; Linköpings universitet; []
    Keywords : MEDICINE; MEDICIN;

    Abstract : Leukotrienes (LTs) are polyunsaturated fatty acid derivatives formed by oxygenation of arachidonic acid via the 5-lipoxygenase (5-LO) pathway. Upon activation of inflammatory cells 5-LO is translocated to the nuclear envelope where it converts arachidonic acid to the unstable epoxide LTA4. READ MORE

  2. 2. Membrane Protein Tailoring and Structural Studies of Leukotriene C4 Synthase

    Author : Daniel Martinez Molina; Pär Nordlund; Robert M. Stroud; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; membrane proteins; random mutagenesis; detergent screening; Leukotriene C₄ synthase; IMAC; Structural biology; Strukturbiologi; Biochemistry; biokemi;

    Abstract : Despite a dramatic increase in the number of proteins that have been structurally characterized in recent years, there are still less than 200 unique structures of membrane proteins known today. This is only 1% of the total number of unique protein structures found in structural databases worldwide. READ MORE

  3. 3. Studies of leukotriene C4 synthase expression and regulation in chronic myeloid leukaemia

    Author : Cecilia Roos; Susanne Tornhamre; Anita Sjölander; Karlstads universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; leukotriene C4 synthase; chronic myeloid leukaemia; p210 BCR-ABL; myelopoiesis; imatinib mesylate; Cell biology; Cellbiologi; Biomedical Sciences; Biomedicinsk vetenskap;

    Abstract : Leukotriene (LT) C4 is generated through conjugation of LTA4 with glutathione by the actions of leukotriene C4 synthase (LTC4S). This biologically potent mediator of asthma and inflammation has also been suggested to play a regulatory role in human myelopoiesis and is overproduced by blood and bone marrow cells from patients with chronic myeloid leukaemia (CML). 4. 4. Mechanism of enzymes involved in leukotriene C4 biosynthesis

    Author : H R Shabbir Ahmad; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Cysteinyl leukotrienes (cys-LTs) are potent proinflammatory mediators associated with various diseases including asthma and allergic rhinitis. Leukotriene C4 synthase (LTC4S) and microsomal glutathione transferase 2 (MGST2) catalyze conjugation of the epoxide intermediate LTA4 with GSH to form LTC4, the parent compound of the cys-LTs. 5. 5. Studies on the formation of leukotriene C4 and lipoxins from leukotriene A4 in human platelets

    Author : Susanne Tornhamre; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Human platelets lack 5-lipoxygenase activity, but possess enzymatic capacity to transform the unstable epoxide leukotriene (LT)A4 to the potent inflammatory agent LTC4 and lipoxins, a group of trihydroxylated tetraene-containing eicosanoids, which recently have been suggested to have an anti-inflammatory role. A novel pathway for the formation of lipoxin (LX)A4 via platelet lipoxygenation of SS,6R-diHETE and 5S,6S-diHETE, enzymatic/non-enzymatic hydrolysis products of LTA4, was discovered in human platelets.
