  1. 1. Intracellular vesicles induced by monotopic membrane protein in Escherichia coli

    University dissertation from Stockholm : Department of Biochemistry and Biophysics, Stockholm University

    Author : Hanna M. Eriksson; Stockholms universitet.; [2009]
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Membrane protein; intracellular vesicles; Escherichia coli; glycosyltransferase; overexpression; optimization; detergent; screening; lipid composition; NATURAL SCIENCES Chemistry Biochemistry; NATURVETENSKAP Kemi Biokemi; biokemi; Biochemistry;

    Abstract : The monotopic membrane protein alMGS, a glycosyltransferase catalyzing glucolipid synthesis in Acholeplasma laidlawii, was overexpressed in Escherichia coli. Optimization of basic growth parameters was performed, and a novel method for detergent and buffer screening using a small size-exclusion chromatography was developed. READ MORE

  2. 2. Structure-Function Relationships of Pi Class Glutathione Transferase Studied by Protein Engineering

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Usama M. Hegazy; Uppsala universitet.; [2006]
    Keywords : Biochemistry; Glutathione transferase; targeted chemical modification; lock-and-key motif; cooperativity; stucture-function relationship; protein engineering; unnatural amino acid; site-specific mutation; Biokemi;

    Abstract : The glutathione transferases (GSTs) represent a superfamily of dimeric proteins involved in cellular detoxication by catalyzing the nucleophilic addition of the reduced glutathione (GSH) to the hydrophobic electrophiles. The present work focuses on the functional role of the conserved structures of GSTP1-1. READ MORE

  3. 3. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Jenny M. Pedersen; Uppsala universitet.; [2013]

    Abstract : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 4. 4. Structure determination and thermodynamic stabilization of an engineered protein-protein complex

    University dissertation from Stockholm : KTH

    Author : Elisabet Wahlberg; KTH.; [2006]
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; affibody; protein structure; coupled folding; NMR spectroscopy; protein stability; protein-protein interactions; binding thermodynamics; isothermal titration calorimetry; protein engineering; TECHNOLOGY Bioengineering Structural biochemistry; TEKNIKVETENSKAP Bioteknik Strukturbiokemi;

    Abstract : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain. 5. 5. The Role of Protein Kinase C in the Extracellular Ca2+-regulated Secretion of Parathyroid Hormone

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Amos M. Sakwe; Uppsala universitet.; [2004]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Medical sciences; parathyroid hormone; Ca2 ; protein kinase C; calcium sensing receptor; ERK1 2; biosynthesis; secretion; MEDICIN OCH VÅRD; MEDICINE; MEDICIN;

    Abstract : Parathyroid hormone (PTH) is the major physiological regulator of the extracellular Ca2+ concentration ([Ca2+]o) in the body. The secretion of this hormone is suppressed at high [Ca2+]o. READ MORE
