  1. 6. Antibody based plasma protein profiling

    Author : Ulrika Qundos; Jochen M. Schwenk; Peter Nilsson; Mathias Uhlén; Ulrike Korf; KTH; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; protein profiling; plasma; antibody; affinity proteomics; biomark- ers; multiplex; assay development;

    Abstract : This thesis is about protein profiling in serum and plasma using antibody suspension bead arrays for the analysis of biobanked samples and in the context of prostate cancer biomarker discovery. The influence of sample preparation methods on antibody based protein profiles were investigated (Papers I-III) and a prostate cancer candidate biomarker identified and verified (Papers III-V). 2. 7. Intracellular vesicles induced by monotopic membrane protein in Escherichia coli

    Author : Hanna M. Eriksson; Åke Wieslander; Meta Kuehn; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Membrane protein; intracellular vesicles; Escherichia coli; glycosyltransferase; overexpression; optimization; detergent; screening; lipid composition; Biochemistry; Biokemi; Biochemistry; biokemi;

    Abstract : The monotopic membrane protein alMGS, a glycosyltransferase catalyzing glucolipid synthesis in Acholeplasma laidlawii, was overexpressed in Escherichia coli. Optimization of basic growth parameters was performed, and a novel method for detergent and buffer screening using a small size-exclusion chromatography was developed. READ MORE

  3. 8. Structure-Function Relationships of Pi Class Glutathione Transferase Studied by Protein Engineering

    Author : Usama M. Hegazy; Bengt Mannervik; Mikael Widersten; T. J. Mantle; Uppsala universitet; []
    Keywords : Biochemistry; Glutathione transferase; targeted chemical modification; lock-and-key motif; cooperativity; stucture-function relationship; protein engineering; unnatural amino acid; site-specific mutation; Biokemi;

    Abstract : The glutathione transferases (GSTs) represent a superfamily of dimeric proteins involved in cellular detoxication by catalyzing the nucleophilic addition of the reduced glutathione (GSH) to the hydrophobic electrophiles. The present work focuses on the functional role of the conserved structures of GSTP1-1. READ MORE

  4. 9. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury

    Author : Jenny M. Pedersen; Per Artursson; Christel A. S. Bergström; Stefan Oswald; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ABC transport protein; Pgp; P-glycoprotein; ABCB1; BCRP; breast cancer resistance protein; ABCG2; MRP2; multidrug resistance-associated protein 2; ABCC2; BSEP; bile salt export pump; ABCB11; sandwich cultured human hepatocytes; SCHH; drug-induced liver injury; DILI; multivariate data analysis; OPLS; Galenisk farmaci; Pharmaceutics;

    Abstract : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 5. 10. Membrane-bound proteins : Characterization, evolution, and functional analysis

    Author : Misty M. Attwood; Helgi B. Schiöth; Arne Elofsson; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Transmembrane protein; alpha-helical membrane protein; evolution membrane proteins; 3TM; 4TM; 5TM; trispanin; Bioinformatics; Bioinformatik;

    Abstract : Alpha-helical transmembrane proteins are important components of many essential cell processes including signal transduction, transport of molecules across membranes, protein and membrane trafficking, and structural and adhesion activities, amongst others. Their involvement in critical networks makes them the focus of interest in investigating disease pathways, as candidate drug targets, and in evolutionary analyses to identify homologous protein families and possible functional activities.
