  1. 1. Inhibition of the MDM2/p53 Interaction Design, Synthesis and Evaluation of MDM2 Inhibitors

    Author : Mariell Pettersson; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Protein-protein interactionProtein-protein interaction; MDM2 p53 interaction; MDM2 inhibitors; α- helix mimetics; β-hairpin; 2; 5-diketopiperazine; spiro-2; 5-diketopiperazine; purine; 8- triazolyl purine; solution conformational analysis; NAMFIS; fluorescence;

    Abstract : Numerous essential cellular processes are regulated by protein-protein interactions (PPIs) and PPIs have therefore been recognised as potential new drug targets. The transcription factor p53 is often referred to as the guardian of the genome due to its involvement in DNA repair, induction of cell cycle arrest and cellular apoptosis. READ MORE

  2. 2. Mdm2 phosphorylations : characterization and applications

    Author : Maria Malmlöf; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Cancer is a widespread disease. In spite of all research resulting in new improved therapies, there are still many tumors that cannot be cured. To oppose that, also a preventive research approach is important with goals such as decreasing the human exposure to carcinogens. READ MORE

  3. 3. Xenobiotics-induced phosphorylations of MDM2

    Author : Gerd Pääjärvi; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The aim of the present study was to characterize the Mdm2 and p53 responses induced by DNA damaging xenobiotics and how these responses can be modified by non-genotoxic xenobiotics. During control conditions cellular levels of p53 are low. The level is regulated by Mdm2 and Mdm2 and p53 forms an autoregulatory loop. READ MORE

  4. 4. Studies of Fusion Oncogenes and Genomic Imbalances in Human Tumors

    Author : Fredrik Persson; Göteborgs universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cancer genetics; fusion oncogene; gene amplification; array CGH; PLAG1; HMGA2; FGFR1; MDM2; pleomorphic adenoma; carcinoma ex pleomorphic adenoma; liposarcoma;

    Abstract : Cancer is a genetic disease caused by mutations and chromosome rearrangements affecting oncogenes and tumor suppressor genes in particular. Molecular analyses of recurrent translocations in hematological disorders, as well as in certain solid tumor types, have shown that they frequently result in fusion oncogenes. READ MORE

  5. 5. Evolution of Interactions Involving Intrinsically Disordered Proteins

    Author : Emma Åberg; Per Jemth; Madan Babu; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; intrinsically disordered proteins; protein evolution; phylogenetics; ancestral sequence reconstruction; biophysical characterization; NCBD; CID; p53; MDM2; Biologi med inriktning mot molekylär evolution; Biology with specialization in Molecular Evolution; Biokemi; Biochemistry;

    Abstract : This thesis describes the evolution of intrinsically disordered proteins and their interaction partners. The work presented is a combination of phylogenetic analysis, ancestral reconstruction and biophysical characterization in order to examine the evolutionary trajectory of protein-protein interactions involving disorder.
