    Abstract : Cyclophosphamide (CPA) is widely used anti-cancer drug that has to be catalyzed by the cytochrome P450 (CYP) enzyme 2B6 as well as some other CYPs to exert its cytotoxic activity. A high degree of interindividual variation in CPA kinetics has been reported in both children and adult patients, which may be due to several factors such as concomitantly used drugs, variation in physiological status of individual patients and genetic polymorphism of the CYP enzymes.     Abstract : Treatment failure in refractory or relapsed disease is often caused by resistance towards Ara-C or daunorubicin, the two drugs mainly used in the treatment of acute myeloid leukemia (AML). This thesis comprises studies of resistance mechanisms regarding Ara-C and the newer nucleoside analogues CdA and fludarabine, improvement of treatment outcome after enhancement of tumour AraC susceptibility by concomitant treatment with GM-CSF and the effect of liposomal formulation on leukemic cell uptake of daunorubicin.
