  1. 6. The non-human primate as a model of human parkinsonism

    Author : Alison McCormack; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Parkinson s disease; substantia nigra; aging; neuromelanin; MPTP; alpha-synuclein; levodopa; dyskinesia; methamphetamine;

    Abstract : The motor deficits seen in Parkinson’s disease (PD) are the result of a loss of catecholaminergic neurons from the substantia nigra (SN) and the associated loss of striatal dopamine. Clinical signs become evident once striatal dopamine levels decline by at least 70-80% and nigral cell numbers by 40%, and worsen with increased duration of the disease. READ MORE

  2. 7. Neurorestorative strategies involving neurogenesis, neuronal precursors and stem cells in animal models of Parkinson's disease

    Author : Ming Zhao; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Parkinson s disease; substantia nigra pars compacta; adult neurogenesis; stereology; apoptosis; dopamine; tyrosine hydroxylase; nestin; 5-bromodeoxyuridine; [3H]-thymidine; MPTP; 6-OHDA; platelet-derived growth factor-BB;

    Abstract : The general aims of the thesis is to provide evidence for neurogenesis in the adult substantia nigra pars compacta (SNpc), where the dopamine-producing neurons lost in Parkinson's disease (PD) reside; to optimize methods used for detecting newborn nerve cells in adult brain regions with a low rate of neurogenesis; to explore the mechanism of nerve cell death in animal models of parkinsonism and early degenerative and restorative changes in nigral cell populations; and to develop a therapeutic approach that could translate into a future restorative disease-modifying strategy in PD. 3. 8. Selective Retention of β-Carbolines and 7,12-Dimethylbenz[a]anthracene in the Brain : Role of Neuromelanin and Cytochrome P450 for Toxicity

    Author : Anna Östergren; Eva Brittebo; Nils Gunnar Lindquist; Anne-Lie Svensson; Hans Tjälve; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Toxicology; β-carboline; neuromelanin; norharman; harman; parkinsonism; Parkinson s disease; motoric impairment; behaviour; glia cells; substantia nigra; dopamine melanin; PC12 cells; ER stress; grp78; hsp90; cytochrome P450; CYP1A1; CYP1B1; blood-brain interfaces; 7; 12-dimethylbenz[a]anthracene; polycyclic aromatic hydrocarbon; endothelial cell; smooth muscle cell; bioactivation; Toxikologi; Toxicology; Toxikologi; Toxicology; toxikologi;

    Abstract : The ß-carbolines norharman and harman structurally resemble the synthetic compound 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) that is known for its ability to damage neuromelanin-containing dopaminergic neurons of the substantia nigra and thereby induce parkinsonism. MPTP is, however, not normally present in the environment whereas the ß-carbolines are present in cooked food and tobacco smoke. READ MORE

  4. 9. Neuropeptidomics – Expanding Proteomics Downwards

    Author : Marcus Svensson; Per Andrén; Per Svenningsson; Jonathan Sweedler; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical pharmacology; proteomics; peptidomics; mass spectrometry; neuropeptide; chromatography; Farmaceutisk farmakologi; PHARMACY; FARMACI;

    Abstract : Biological function is mainly carried out by a dynamic population of proteins which may be used as markers for disease diagnosis, prognosis, and as a guide for effective treatment. In analogy to genomics, the study of proteins is called proteomics and it is generally performed by two-dimensional gel electrophoresis and mass spectrometric methods. READ MORE

  5. 10. Mechanisms of Ischemic Brain Injury- studies in murine hippocampal slice cultures

    Author : Anna Rytter; Sektion IV; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neurophysiology; Neurologi; neuropsykologi; neuropsychology; Neurology; adenosine; caspases; hypothermia; in vitro; hippocampus; ischemia; neurofysiologi;

    Abstract : Cerebral ischemia is a major cause of mortality and morbidity in the western world. Even though much effort has been put into research and development of effective drugs against ischemic damage there is to date no effective pharmacological brain protective treatment. READ MORE
