  1. 1. Methodological Studies on Models and Methods for Mixed-Effects Categorical Data Analysis

    Author : Maria C. Kjellsson; Mats O. Karlsson; Siv Jönsson; Gordon Graham; Uppsala universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacodynamics; Categorical data; Markov model; Modelling; NONMEM; NLMIXED; Laplace; Gaussian quadrature; Back-Step Method; Proportional odds model; Differential odds model;

    Abstract : Effects of drugs are in clinical trials often measured on categorical scales. These measurements are increasingly being analyzed using mixed-effects logistic regression. However, the experience with such analyzes is limited and only a few models are used. READ MORE

  2. 2. Mechanism-Based Modelling of Clinical and Preclinical Studies of Glucose Homeostasis

    Author : Oskar Alskär; Maria C Kjellsson; Mats O Karlsson; Jenny Y. Chien; Uppsala universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; glucose homeostasis; pharmacometrics; Farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy;

    Abstract : Glucose is an important nutrient and energy source in the body. However, too high concentration in the blood is harmful and may lead to several complications developing over time. It was estimated that 5 million people in the world died from complications related to diabetes during 2015. READ MORE

  3. 3. Pharmacometrics Modelling in Type 2 Diabetes Mellitus : Implications on Study Design and Diabetes Disease Progression

    Abstract : Pharmacometric modelling is widely used in many aspects related to type 2 diabetes mellitus (T2DM), for instance in the anti-diabetes drug development, and in quantifying the disease progression of T2DM.The aim of this thesis were to improve the design of early phase anti-diabetes drug development studies with the focus on the power to identify mechanism of drug action (MoA), and to characterize and quantify the progression from prediabetes to overt diabetes, both the natural progression and the progression with diet and exercise interventions, using pharmacometrics modelling. 4. 4. Pharmacometric evaluation and improvement of models and study designs - applied in diabetes

    Author : Moustafa M. A. Ibrahim; Mats O. Karlsson; Maria C. Kjellsson; Paolo Magni; Uppsala universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES;

    Abstract : Pharmacometric models are increasingly used to improve the efficiency of the drug development process and increase our understanding of the studied underlying pathophysiological system. These models require assumptions for handling different types of data and the different model components, and the appropriateness of such assumptions must be carefully inspected for unbiased conclusions. 5. 5. Pharmacometric Models to Improve Treatment of Tuberculosis

    Author : Elin M Svensson; Mats O Karlsson; Maria C Kjellsson; Ulrika S H Simonsson; N. L’Ntshotsholé (Shasha) Jumbe; Uppsala universitet; []
    Keywords : pharmacokinetics; pharmacodynamics; population approach; nonlinear mixed-effects models; multidrug-resistant tuberculosis; bedaquiline; antiretroviral; drug-drug interactions; time-to-event; albumin; Klinisk farmakologi; Clinical Pharmacology;

    Abstract : Tuberculosis (TB) is the world’s most deadly infectious disease and causes enormous public health problems. The comorbidity with HIV and the rise of multidrug-resistant TB strains impede successful therapy through drug-drug interactions and the lack of efficient second-line treatments. READ MORE
