  1. 11. Redefining the essential molecular aspects that drive interactions between small molecules and G-quadruplex DNA

    Author : Måns Andréasson; Erik Chorell; Anna Linusson Jonsson; Jan Kihlberg; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; G-Quadruplexes; DNA; Oncogenes; G4 ligands; heterocycles; macrocycles; organic synthesis; molecular interactions; rational compound design; medicinal chemistry.; medicinal chemistry; läkemedelskemi; organisk kemi; Organic Chemistry;

    Abstract : G-Quadruplex (G4) structures are secondary nucleic acid structures located in guanine-rich regions of DNA and RNA sequences, involved in gene regulation and cellular maintenance. Efforts to target G4s in a therapeutic setting are scarce, mainly due to vague details about the binding interactions between the ligands and the G4 structure combined with the lack of emphasis on drug-like properties early in the ligand development process. 2. 12. Design and Synthesis of Acyclic and Macrocyclic Peptidomimetics as Inhibitors of the Hepatitis C Virus NS3 Protease

    Author : Anna Lampa; Anja Sandström; Youla Tsantrizos; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; hepatitis C virus; HCV; NS3 protease inhibitor; structure-activity relationship; phenylglycine; ring-closing metathesis; Medicinal Chemistry; Läkemedelskemi;

    Abstract : Hepatitis C is a blood-borne disease affecting 130-170 million people worldwide. The causative agent, hepatitis C virus (HCV), infects the liver and is the major reason for chronic liver disease worldwide. The HCV NS3 protease, a key enzyme in the virus replication cycle, has been confirmed to be an important target for drug development. READ MORE

  3. 13. New Methods for the Synthesis of 3-Substituted 1-Indanones : A Palladium-Catalyzed Approach

    Author : Anna Arefalk; Anders Hallberg; Mats Larhed; Juan Carlos Carretero; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; Heck; palladium; indanone; annulation; microwave heating; triflate; sulfinyl imine; HIV-1 protease inhibitor; Organisk kemi; Organic chemistry; Organisk kemi;

    Abstract : In medicinal chemistry, there is a constant need for new preparative methods, both to make the synthesis process more effective, and to increase the accessibility to a wide variety of compounds. A number of different approaches can be used to attain these goals. READ MORE

  4. 14. C1 Building Blocks: New Approaches for Thiomethylations and Esterification

    Author : Marcus Söderström; Luke R. Odell; Anders Karlén; Vittorio Pace; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Borontrifluoride; Dimethylsulfide; Thiomethylation; Methylthioethers; Dithioacetalation; Methyl-Dithioacetals; Friedel-Crafts; Diarylmethanes; Defluorination; Methyl-Dithioesters; Thioamides; Diazomethane; Esterification; Medicinal Chemistry; Läkemedelskemi; Medicinal Chemistry; Läkemedelskemi;

    Abstract : The thioether motif is found in numerous pharmaceuticals. The simplest form of this motif, the methyl-thioether is similarly found in many biologically active compounds and has exhibited many advantageous properties. The installation of the thiomethyl moiety can be troublesome due to the toxicity and malodor of small sulfur reagents. READ MORE

  5. 15. Palladium(II)-Catalyzed Heck Reactions : Domino Reactions, Decarboxylations, Mechanistic Studies & Continuous Flow Applications

    Author : Ashkan Fardost; Mats Larhed; Patrick Guiry; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; palladium II ; dft; reaction mechanisms; esi-ms; heck; decarboxylation; microwave; continuous flow;

    Abstract : This thesis describes research efforts dedicated to the development of palladium(II)-catalyzed oxidative Heck and Heck/Suzuki domino reactions, and the applications of a new microwave heating technology, purpose-built for continuous flow in organic synthesis.Paper I describes the development of a ligand-modulated approach for attaching aryl groups to a chelating vinyl ether.
