  1. 16. Design and Synthesis of Acyclic and Macrocyclic Peptidomimetics as Inhibitors of the Hepatitis C Virus NS3 Protease

    Author : Anna Lampa; Anja Sandström; Youla Tsantrizos; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; hepatitis C virus; HCV; NS3 protease inhibitor; structure-activity relationship; phenylglycine; ring-closing metathesis; Medicinal Chemistry; Läkemedelskemi;

    Abstract : Hepatitis C is a blood-borne disease affecting 130-170 million people worldwide. The causative agent, hepatitis C virus (HCV), infects the liver and is the major reason for chronic liver disease worldwide. The HCV NS3 protease, a key enzyme in the virus replication cycle, has been confirmed to be an important target for drug development. READ MORE

  2. 17. Design and Synthesis of Hepatitis C Virus NS3 Protease Inhibitors

    Author : Anja Johansson; Montse Llinàs-Brunet; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical chemistry; Farmaceutisk kemi; Pharmaceutical chemistry; Farmaceutisk kemi; Medicinal Chemistry; Läkemedelskemi;

    Abstract : Hepatitis C Virus (HCV) is the leading cause of chronic liver disease worldwide as well as the primary indication for liver transplantation. More than 3% of the world’s population is chronically infected with HCV and there is an urgent need for effective therapy. READ MORE

  3. 18. Kinetic studies of NS3 and NS5B from Hepatitis C virus : Implications and applications for drug discovery

    Author : Göran Dahl; Helena Danielson; Raffaele De Francesco; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Hepatitis C virus; NS3; NS5B; enzyme kinetics; inhibition; resistance; drug; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : The aim of these studies was to increase our understanding of the non-structural proteins 3 and 5B (NS3 and NS5B) from the hepatitis C virus (HCV), and thereby contribute to the development of new and better drugs against HCV.By studying NS3 with substitutions identified to be associated with resistance to NS3 inhibitors in clinical trials (R155Q, A156T and D168V) it was found that not all inhibitors were affected, indicating that cross-resistance can be avoided. 4. 19. Design and Synthesis of Enzyme Inhibitors Against Infectious Diseases : Targeting Hepatitis C Virus NS3 Protease and Mycobacterium tuberculosis Ribonucleotide Reductase

    Author : Johan Gising; Mats Larhed; Anders Karlén; Victor Snieckus; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Medicinal Chemistry; Läkemedelskemi;

    Abstract : Infectious diseases, including hepatitis C and tuberculosis, claim the lives of over 15 million people each year. Hepatitis C is caused by the hepatitis C virus (HCV) which infects the liver and can ultimately result in liver transplantation. HCV is very adaptive as a result of its high mutation rate. READ MORE

  5. 20. Interaction Characteristics of Viral Protease Targets and Inhibitors : Perspectives for drug discovery and development of model systems

    Author : Cynthia F Shuman; Jordan J N Tang; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; viral proteases; biomolecular interactions; kinetics; thermodynamics; biosensors; drug discovery; Biokemi; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : Viral proteases are important targets for anti-viral drugs. Discovery of protease inhibitors as anti-viral drugs is aided by an understanding of the interactions between viral protease and inhibitors. This thesis addresses the characterization of protease-inhibitor interactions for application to drug discovery and model system development. READ MORE
