  1. 16. PET and the Multitracer Concept: An Approach to Neuroimaging Pathology

    Author : Henry Engler; Bengt Långström; Göran Hedenstierna; Juha Rinne; Uppsala universitet; []
    Keywords : Neurosciences; PET Positron Emission Tomography ; multitracer; neuroimaging pathology; astrocytes; microglia; neurodegeneration; amyloid; AD; CJD; PD; Neurovetenskap;

    Abstract : Patients suffering from different forms of neurodegenerative diseases, such as: Creutzfeldt Jacob Disease (CJD), Alzheimer disease (AD), mild cognitive impairment (MCI), frontotemporal dementia and Parkinson’s disease (PD) were examined with Positron Emission Tomography (PET) and the combination of different radiotracers: 15O-water, N-[11C-methyl]-L-deuterodeprenyl (DED), [18F] 2-fluorodeoxyglucose: (FDG), N-methyl-[11C]2-(4-methylaminophenyl)-6-hydroxybenzothiazole (PIB) and L-[11C]-3,4-dihydroxiphenyl-alanine (DOPA). The radiotracers and the combinations of different radiotracers were selected with the intention to detect, in the brain, patterns of neuronal dysfunction, astrocytosis, axon degeneration or protein aggregation (amyloid), in the brain which are pathognomonic for specific diseases and may contribute to improve clinical differential diagnoses. 2. 17. A Drosophila Disease-Model for Transthyretin-associated Amyloidosis

    Author : Malgorzata Pokrzywa; Erik Lundgren; Damian Crowther; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; transthyretin; amyloid; amyloidosis; Drosophila; transthyretin-associated amyloidosis; Familial amyloid polyneuropathy; neurodegeneration; serum amyloid P component; Non-steroidal Anti-Inflammatory Drugs; drug screens; Molecular biology; Molekylärbiologi;

    Abstract : Amyloidoses comprise a group of gain-of-toxic function protein misfolding diseases, in which normally soluble proteins in their functional state undergo conformational changes into highly organized and generally intractable thread-like aggregates, termed amyloid fibrils. These structures accumulate predominantly in the extracellular space but growing evidence suggests that amyloids may start to form intracellularly. READ MORE

  3. 18. The future of clinical trials for Alzheimer's disease. A blood-based biomarker perspective

    Author : Nicholas Cullen; Klinisk minnesforskning; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer s disease; Biomarkers; Cognition; Amyloid; Tau; Neurodegeneration;

    Abstract : Objectives: The primary objective was to investigate the utility of blood-basedbiomarkers of amyloid, tau, and neurodegeneration for (i) screening, (ii)enrichment, and (iii) tracking response to treatment in clinical trials of Alzheimer’sdisease.Methods: Longitudinal, participant-level data used in these studies was drawn fromthe Swedish BioFINDER study and the ADNI study. 4. 19. A stargazer's guide to neurodegeneration : Astrocytes' role in the propagation of pathological proteins

    Author : Tobias Mothes; Anna Erlandsson; Martin Ingelsson; Linn Streubel-Gallasch; Maria Grazia Spillantini; Uppsala universitet; []
    Keywords : Alzheimer’s disease; Parkinson’s disease; tauopathies; astrocyte; microglia; neuron; tau; amyloid-β; α-synuclein; accumulation; degradation; inflammation; spreading; seeding.; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract : Alzheimer’s disease (AD) and Parkinson’s disease (PD) are characterized by brain accumulation of pathogenic protein aggregates. In the AD brain, amyloid-β (Aβ) and tau form plaques respective tangles, while in the PD brain α-synuclein (α-syn) form Lewy bodies and Lewy neurites. READ MORE

  5. 20. β-cell dysfunction in Huntington's disease

    Author : Karl Bacos; Institutionen för experimentell medicinsk vetenskap; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; metabolism; secretion; Insulin; trafficking; R6 2 mouse; granule; exocytosis; neurodegeneration; huntingtin;

    Abstract : Huntington's disease (HD), a fatal neurodegenerative disorder, is associated with an increased risk of diabetes mellitus. The reason for this is unknown, but considering the functional similarities of neurons and the insulin secreting pancreatic β-cell, pathological mechanisms may be shared by both cell types and account for neuronal as well as endocrine dysfunction.
