  1. 11. In Vivo Active Drug Uptake and Efflux at the Blood-Brain Barrier : With Focus on Drug Transport Interactions

    Author : Muhammad Waqas Sadiq; Margareta Hammarlund-Udenaes; Sven Björkman; N. Joan Abbott; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Blood-brain barrier; Active uptake transport; Microdialysis; P-glycoprotein; Drug interactions; Species differences; Pharmacokinetics; Pharmacodynamics; NONMEM; Pharmaceutical Science; Farmaceutisk vetenskap;

    Abstract : The blood-brain barrier (BBB) controls the movement of substances into and out of the brain. The tight junctions between endothelial cells and energy dependent transporters in the BBB influence rate and extent of drug distribution to the brain. READ MORE

  2. 12. Clinical studies of CYP3A4 and P-glycoprotein

    Author : Erik Sparve; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : In the field of pharmacokinetics, cytochrome P450 enzymes have been extensively studied. The CYP450 enzyme with the most drug substrates is CYP3A4, but despite its significance, administration of a specific substrate to subjects is required to determine its activity, by measuring the metabolic ratio. 3. 13. Pharmacokinetics and Pharmacodynamics of Oxycodone and Morphine with Emphasis on Blood-Brain Barrier Transport

    Author : Emma Boström; Margareta Hammarlund-Udenaes; Ulrika Simonsson; Danny D Shen; Uppsala universitet; []
    Keywords : Pharmacokinetics Pharmacotherapy; pharmacokinetics; pharamcodynamics; blood-brain barrier; oxycodone; microdialysis; NONMEM; brain distribution; transport; Farmakokinetik Farmakoterapi;

    Abstract : The pharmacokinetics and pharmacodynamics of oxycodone and morphine was investigated and related to the transport across the blood-brain barrier (BBB) in rats. The influence of a P-glycoprotein (P-gp) inhibitor on the plasma pharmacokinetics and pharmacodynamics of oxycodone was evaluated. READ MORE

  4. 14. Intracellular unbound drug concentrations : Methodology and application for understanding cellular drug exposure

    Author : André Mateus; Per Artursson; Pär Matsson; José A. G. Morais; Jeff Krise; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intracellular unbound drug concentrations; free drug; drug binding; drug transport; drug accumulation; cellular drug response; drug target engagement; Pharmaceutical Science; Farmaceutisk vetenskap;

    Abstract : Most known drug targets and metabolizing enzymes are located inside cells. Interactions with these proteins are determined by intracellular unbound drug concentrations. Assessing intracellular drug exposure is technically challenging, but essential for predicting pharmacokinetic, pharmacological, and toxicological profiles of new drugs. READ MORE

  5. 15. Pharmacological and clinical studies of new ways to improve cytostatic treatment of acute myelocytic leukemia : in vitro and in vivo studies

    Author : Christina Löfgren; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Treatment failure in refractory or relapsed disease is often caused by resistance towards Ara-C or daunorubicin, the two drugs mainly used in the treatment of acute myeloid leukemia (AML). This thesis comprises studies of resistance mechanisms regarding Ara-C and the newer nucleoside analogues CdA and fludarabine, improvement of treatment outcome after enhancement of tumour AraC susceptibility by concomitant treatment with GM-CSF and the effect of liposomal formulation on leukemic cell uptake of daunorubicin.
