  1. 1. Zebrafish models for large-scale genetic screens in dyslipidemia and atherosclerosis : Validation and application

    Author : Manoj Kumar Bandaru; Marcel den Hoed; Anastasia Emmanouilidou; Ingelsson Erik; Shannon Fisher; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; zebrafish; dyslipidemia; atherosclerosis; genetic screens; high-cholesterol diet; APOE; APOB; LDLR; APOC2; PCSK9; LPAR2; GATAD2A; Medicinsk genetik; Medical Genetics;

    Abstract : Hundreds of loci have been robustly associated with circulating lipids, atherosclerosis and coronary artery disease; but for most loci the causal genes and mechanisms remain uncharacterized. The overall aim of my thesis is to develop and validate novel in vivo model systems that are suitable for high-throughput, image-based genetic screens in coronary artery disease and related traits, and use these model systems to systematically characterize positional candidate genes. 2. 2. The role of HMG-coenzyme A reductase (HMGCR) and statin medication in the Central Nervous System : Cognitive Functions, Metabolism, Feeding and Sleep Behaviour

    Author : Ahmed Alsehli; Helgi B. Schiöth; Michael J. Williams; Heinrich Dircksen; Uppsala universitet; []
    Keywords : Statin; cardiovascular disease; HMGCR; PCSK9; sleep; insomnia; circadian; Chronotype; feeding;

    Abstract : Millions of people are currently on statin medications (HMGCR inhibitors) to prevent cardiovascular diseases. Despite considerable central nervous system expression, little is known about HMGCR function in the brain. READ MORE

  3. 3. Studies on PCSK9 in the regulation of cholesterol metabolism

    Author : Lena Persson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : aging;

    Abstract : Elevated levels of plasma cholesterol, mainly in low density lipoproteins (LDL), are a major risk factor for coronary heart disease. The level of plasma LDL cholesterol (LDL-C) is largely dependent on the number of hepatic LDL receptors (LDLRs). READ MORE

  4. 4. Genetic and metabolic studies of APOE, PCSK9, TM6SF2 and PNPLA3

    Author : Hovsep Mahdessian; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Lipids – including fatty acids (FAs), triglycerides (TGs), cholesterol and cholesterol esters (CEs) – are hydrophobic molecules involved in several important structural and mechanistic processes. Lipids are found in cellular membranes, give posture and stability to the cell, are actively participating in cellular signaling, and act as mediators in several important biological pathways. 5. 5. Hormonal influences on cholesterol and bile acid metabolism : focusing on involvement of PCSK9

    Author : Moumita Ghosh Laskar; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Elevated plasma LDL-C is today considered a causative factor for the development of atherosclerosis. Increased plasma LDL-C levels are mainly due to reduced numbers of functional LDL receptors (LDLRs) in the liver. READ MORE
