  1. 1. Models for the Transfer of Drugs from the Nasal Cavity to the Central Nervous System

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Björn Jansson; Uppsala universitet.; [2004]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biopharmacy; Administration; intranasal; Nasal mucosa; Central nervous system; Olfactory bulb; Rat; Mouse; Swine; Cattle; Diffusion chambers; culture; In vitro; Biological models; Biological transport; Dextrans; Dopamine; Gels; Polysaccharides; Autoradiography; Fluorescence microscopy; Biofarmaci; PHARMACY Biopharmacy; FARMACI Biofarmaci;

    Abstract : The blood-brain barrier restricts the access of many compounds, including therapeutic agents, to the brain. Several human studies indicate that nasal administration of hydrophilic compounds, such as peptides, can bypass the blood-brain barrier. The aims of this thesis were to develop and refine models for this direct nose-to-brain transfer. READ MORE

  2. 2. Validation of Several Nucleoside Tracers as Proliferation Markers

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Li Lu; Uppsala universitet.; [2002]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biopharmacy; PET; cell proliferation; DNA synthesis; hydroxyurea; cimetidine; diuretic; Biofarmaci; PHARMACY Biopharmacy; FARMACI Biofarmaci; galenisk farmaci; Pharmaceutics;

    Abstract : Positron emission tomography (PET) is a powerful method for in vivo characterization of tumor biochemistry and clinically has been used for diagnosing and grading of malignancy. A key element in oncology is proliferation potential, a feature that is assumed to be an important indicator of tumor growth. READ MORE

  3. 3. Intestinal Permeability and Presystemic Extraction of Fexofenadine and R/S-verapamil

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Christer Tannergren; Uppsala universitet.; [2004]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biopharmacy; Bioavailability; Fexofenadine; Verapamil; P-glycoprotein; CYP3A4; OATP; Enantioselective; Permeability; Caco-2; Intestinal perfusion; Biofarmaci; PHARMACY Biopharmacy; FARMACI Biofarmaci;

    Abstract : The main objective of this thesis was to investigate the in vivo relevance of membrane transporters and cytochrome P450 (CYP) 3A4-mediated metabolism in the intestine and liver for the bioavailability of drugs in humans after oral administration.In the first part of the thesis, the main transport mechanisms involved in the intestinal absorption and bioavailability were investigated for fexofenadine, a minimally metabolized drug, which is a substrate for P-glycoprotein (P-gp) and members of organic anion transporting polypeptide (OATP) family. 4. 4. Development of Methods for Assessing Unbound Drug Exposure in the Brain In vivo, in vitro and in silico

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Markus Fridén; Uppsala universitet.; [2010]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Blood-brain barrier; Drug transport; Tissue distribution; Transporters; Pharmacokinetics; PHARMACY Other pharmacy; FARMACI Övrig farmaci; PHARMACY Biopharmacy; FARMACI Biofarmaci; PHARMACY Pharmaceutical pharmacology; FARMACI Farmaceutisk farmakologi; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Abstract : The blood-brain barrier is formed by tightly joined capillary cells with transporter proteins and acts as to regulate the brain concentration of nutrients as well as many drugs. When developing central nervous system drugs it is necessary to measure the unbound drug concentration in the brain, i.e. the unbound brain exposure. READ MORE

  5. 5. Involvement of Membrane Transport Proteins in Intestinal Absorption and Hepatic Disposition of Drugs Using Fexofenadine as a Model Drug

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Niclas Petri; Uppsala universitet.; [2005]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biopharmacy; Fexofenadine; OATP; P-glycoprotein; Organic Anion Transporters; ATP-Binding Cassette Transporters; membrane transport proteins; Bioavailability; Biopharmaceutics; Biofarmaci; PHARMACY Biopharmacy; FARMACI Biofarmaci;

    Abstract : The aims of this thesis were to study the in vivo relevance of membrane transporters for intestinal absorption and the hepatic disposition of drugs in humans and preclinical models. Fexofenadine is a substrate for ABCB1 (P-glycoprotein) and members of the organic anion transporting polypeptide (OATP/SLCO) family. READ MORE
