  1. 6. Hepatic Disposition of Drugs and the Utility of Mechanistic Modelling and Simulation

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Erik Sjögren; Uppsala universitet.; [2010]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Hepatic disposition; pharmacokinetics; mechanistic modelling; drug-drug interactions; enzyme kinetics; Vmax; Km; CLint; carrier-mediated transport; active transport; modelling; in vitro-in vivo extrapolation; physiologically based pharmacokinetic model; optimal experimental design; experimental optimization; data analysis optimization; PHARMACY; FARMACI; PHARMACY Biopharmacy; FARMACI Biofarmaci; Biopharmaceutics; Biofarmaci;

    Abstract : The elimination of drugs from the body is in many cases performed by the liver. Much could be gained if an accurate prediction of this process could be made early in the development of new drugs. However, for the elimination to occur, the drug molecule needs first to get inside the liver cell. READ MORE

    Abstract : Drug transport across cells and cell membranes in the human body is crucial for the pharmacological effect of drugs. Active transport governed by transport proteins plays an important role in this process. A vast number of transport proteins with a wide tissue distribution have been identified during the last 15 years. READ MORE

  3. 8. Metabolism-dependent activation of food and environmental mutagens in endothelial cells

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Anita Annas; Uppsala universitet.; [2000]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; Farmaceutisk biovetenskap; PHARMACY Biopharmacy; FARMACI Biofarmaci; Toxicology; toxikologi;

    Abstract : The endothelial cells of blood vessels have been proposed as a target for toxic effects ofxenobiotics in the cardiovascular system. In the present studies, induction of cytochromeP450 1A (CYP1A) enzymes and metabolic activation of food and environmentalmutagens were examined in endothelial cells of rodents, birds, and humans. 4. 9. Cytochrome P450 enzymes in the biosynthesis and metabolism of bile acids and active forms of vitamin D

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Zufan Araya; Uppsala universitet.; [1999]
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; Farmaceutisk biovetenskap; PHARMACY Biopharmacy; FARMACI Biofarmaci; farmaceutisk biokemi; Pharmaceutical Biochemistry;

    Abstract : Cytochrome P450 enzymes catalyzing hydroxylations in the biosynthesis and metabolism of bile acids and active forms of vitamin D3 were studied.A microsomal cytochrome P450 enzyme with a molecular mass of 53 kDa, catalyzing the 6α-hydroxylation of taurochenodeoxycholic acid was purfied from pig liver microsomes and characterized with respect to catalytic and electrophoretic properties as well as polyclonal antibodies. 5. 10. A study on the vascular effects of prostaglandin F2α[ and latanaprost in the eye with special reference to the surface hyperaemia after topical administration

    University dissertation from Uppsala : Acta Universitatis Upsaliensis

    Author : Maria Astin; Uppsala universitet.; [1998]

    Abstract : The ocular hypotensive effect of topically applied prostaglandins is usually accompanied byconjunctival hyperaemia and irritation. The purpose of the present study was to investigate thepharmacological mechanism of the ocular surface hyperaemia induced by topically appliedprostaglandins, particularly prostaglandin F2α (PGF2α) and its synthetic analogue latanoprost, aselective FP prostanoid receptor agonist.
