  1. 1. Chemoenzymatic Resolution in Dynamic Systems : Screening, Classification and Asymmetric Synthesis

    Author : Yan Zhang; Olof Ramström; Sabine Flitsch; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; constitutional dynamic chemistry; dynamic systemic resolution; dynamic kinetic resolution; enzyme catalysis; transesterification; enzyme promiscuity; asymmetric synthesis; pattern recognition; self-inhibition.;

    Abstract : This  thesis  is  divided  into  four  parts,  all  centered  around  Constitutional Dynamic  Chemistry  (CDC)  and  Dynamic  Kinetic  Resolution  (DKR)  using biocatalysts for selective transformations, and their applications in screening of bioactive compounds, organic synthesis, and enzyme classification.   In  part  one,  an  introduction  to  CDC  and  DKR  is  presented,  illustrating  the basic  concepts,  practical  considerations  and  potential  applications  of  such dynamic systems, thus providing the background information for the studies in the following chapters. 2. 2. Methods for Asymmetric Olefination Reactions; Development and Application to Natural Product Synthesis

    Author : Daniel Strand; Tobias Rein; Viresh Rawal; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Antitumor agents; Asymmetric Horner-Wadsworth-Emmons; Desymmetrization; Metal-catalyzed olefination; Mucocin; Palladium-catalyzed allylic substitution; Parallel kinetic resolution; Pyranicin; Pyragonicin; Rhenium; Stereoconvergent synthesis; Stereodivergent synthesis; Stereoselective synthesis; Tetrahydropyran; Wittig reaction; Organic chemistry; Organisk kemi;

    Abstract : This thesis deals with the development and application of methods for asymmetric olefinations, in particular Horner-Wadsworth-Emmons (HWE) reactions, in the synthesis of certain natural products. Relying on asymmetric HWE reactions to access key building blocks, two natu-ral products, pyranicin and pyragonicin, were synthesized from common late intermediates. READ MORE

  3. 3. Resolution of Dynamic Systems: Applications in Asymmetric Synthesis and Materials Science

    Author : Lei Hu; Olof Ramström; Douglas Philp; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Kemi; Chemistry;

    Abstract : This thesis is built on the concepts of constitutional dynamic chemistry and dynamic kinetic resolution, where reversible covalent reactions are used to generate dynamics and kinetically controlled reactions are employed for resolution. The thesis is divided into two parts:The first section focuses on the study of dynamic systemic resolution, a concept derived from constitutional dynamic chemistry. READ MORE

  4. 4. Reducing Attrition via Improved Strategies for Pre-clinical Drug Discovery : SPR-biosensor Aided Interaction Studies

    Author : Johan Winquist; Helena Danielson; Roderick E. Hubbard; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biokemi; Biochemistry;

    Abstract : The efficacy of a drug is tightly intertwined with its interaction mechanism with the drug target. The mechanism is dependent on the physicochemical and structural characteristics of both target and drug molecule.Drug discovery is plagued by a high attrition rate, whereas in the clinic, a major issue is drug resistance. 5. 5. Biocatalytic transamination with recombinant Saccharomyces cerevisiae: Challenges and opportunities

    Author : Nora Weber; Teknisk mikrobiologi; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Saccharomyces cerevisiae; transamination; whole-cell biocatalysis; pyruvate; PLP; thiamine; reduction;

    Abstract : Chiral building blocks are important molecules for synthesis of fine chemicals and pharmaceuticals. Biocatalysis has gained in relevance over traditional organic methods for synthesis of chiral compounds, as reactions can be performed at high enantio-selectivity and purity with environmentally friendly, simple, and cheap methods.
