  1. 1. Design of solid dosage forms for mucosal vaccination - Investigations on the influence of excipients on product performance

    Author : Annika Borde; Chalmers tekniska högskola; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; hydrophilic matrix tablets; osmotic pressure; protein release; freeze-drying; Vibrio cholerae; mucoadhesion; Bradford Assay; sublingual vaccine tablet; mucosal vaccination;

    Abstract : Most vaccines today are liquid formulations for parental administration. However, there are several drawbacks connected to these vaccines. Since injectable vaccines only induce systemic antibody responses, they are not effective against the various pathogens that affect mucosal surfaces with poor permeability for serum-derived antibodies, e.g. READ MORE

  2. 2. Cell line and protein engineering tools for production and characterization of biologics

    Author : Anna-Luisa Volk; Johan Rockberg; Mathias Uhlen; Dickson Alan; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Pharmaceutical proteins; precision medicine; antibody engineering; epitope mapping; cell line development; split-GFP; Bioteknologi; Biotechnology;

    Abstract : Our increasing understanding of disease mechanisms coupled with technological advances has facilitated the generation of pharmaceutical proteins, which are able to address yet unmet medical needs. Diseases that were fatal in the past can now be treated with novel biological medications improving and prolonging life for many patients. READ MORE

  3. 3. Metabolic Engineering of Recombinant Protein Production by Saccharomyces cerevisiae

    Author : Zihe Liu; Chalmers tekniska högskola; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; recombinant protein production; genome sequencing; DNA microarray; insulin precursor; growth rate; UV mutation; yeast; anaerobic electron acceptor; secretory pathway; unfolded protein response; metabolic engineering; systems biology; α-amylase;

    Abstract : The yeast Saccharomyces cerevisiae is a widely used cell factory for the production of fuels, chemicals, and it also provides a platform for the production of many heterologous proteins of medical or industrial interest. In this thesis, random and rational approaches, such as vector design, host engineering, fermentation analysis, UV Mutation, coupled with high-throughput systems biology techniques (including whole genomic sequencing, microarray analysis and flux analysis) and integrated analysis (Reporter feature technique), were employed to engineer cellular properties more effectively and purposefully to construct cell factories for protein production. 4. 4. Generation and engineering of ABD-derived affinity proteins for clinical applications

    Author : Sarah Lindbo; Sophia Hober; Christoph Rader; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; ABD; ADAPT; affinity proteins; protein engineering; radionuclide molecular imaging; HER2; Bioteknologi; Biotechnology;

    Abstract : Proteins that specifically recognize and bind to other molecules or structures are important tools in industrial and medical applications. Binding proteins engineered from small stable scaffold proteins have been utilized for several purposes due to their favorable biophysical properties, tolerance to mutagenesis, efficient tissue penetration and ease of production. Abstract : Cancer is a major public health challenge and the second leading cause of death in the world, with millions of new cases being diagnosed each year. Traditional cancer treatments such as surgery, radiation therapy, and chemotherapy are many times effective, but may also cause damage to healthy cells, leading to side effects. READ MORE
