  1. 21. Studies of the Neuropeptide Y Receptor Y2 in Human and Zebrafish

    Author : Helena Fällmar; Dan Larhammar; Ingrid Lundell; Nina Mohell; Raquel E. Rodríguez Rodríguez; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Y2-receptor; NPY; PYY; G-protein-coupled receptor; mutagenesis; receptor binding; zebrafish; evolution; Pharmacology; Farmakologi; Farmakologi; Pharmacology;

    Abstract : The G-protein coupled receptors (GPCRs) comprise the largest family of receptors in humans and other vertebrates. They are embedded in the cell membrane and are activated by many different signaling molecules. Activation modulates cellular signal transduction pathways and influences many physiological processes. 2. 22. Catalytic and Structural Properties of Heme-containing Fatty Acid Dioxygenases : Similarities of Fungal Dioxygenases and Cyclooxygenases

    Author : Ulrike Garscha; Ernst Oliw; Per-Johan Jakobsson; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; dioxygenase; prostaglandin H synthase; Gaeumannomyces graminis; Aspergilli; hydroperoxide isomerase; linoleate diol synthase; cytochrome P450; oxylipin; Pharmaceutical pharmacology; Farmaceutisk farmakologi; Farmaceutisk farmakologi; Pharmaceutical Pharmacology;

    Abstract : 7,8-Linoleate diol synthase (7,8-LDS) of the take-all pathogen of wheat, Gaeumannomyces graminis, converts linoleic acid to 8R-hydroperoxyoctadecadienoic acid (8-HPODE) by 8-dioxygenase activity (8-DOX), and further isomerizes the hydroperoxide to 7S,8S-dihydroxyoctadecadienoic acid (7,8-DiHODE) by hydroperoxide isomerase activity. Sequence alignment showed homology to prostaglandin H synthase (PGHS), and both enzymes share structural and catalytic properties. READ MORE

    Abstract : Semicarbazide-sensitive amine oxidase (SSAO, EC 1.4.3.6) belongs to a family of copper-containing amine oxidases. READ MORE

  4. 24. Integrating Efficacy and Toxicity in Preclinical Anticancer Drug Development : Methods and Applications

    Author : Caroline Haglund; Elin Lindhagen; Rolf Larsson; Anna Åleskog; Sigurd Vitols; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Anticancer drugs; In vitro assays; Toxicity testing; Haematological toxicity; Primary tumour cells; Species difference; Clinical pharmacology; Klinisk farmakologi; Clinical Pharmacology; Klinisk farmakologi;

    Abstract : Preclinical testing is an important part of cancer drug development. The aim of this thesis was to establish and evaluate preclinical in vitro methods useful in the development of new anticancer drugs. In paper I, the development of non-clonogenic assays (FMCA-GM) using CD34+ stem cells for assessment of haematological toxicity was described. READ MORE

  5. 25. Methods for Preclinical Evaluation of Cytotoxic Drugs : With Special Reference to the Cyanoguanidine CHS 828 and Hollow Fiber Method

    Author : Saadia Bashir Hassan; Elin Lindhagen; Rolf Larsson; Mats O Karlsson; Mustapha Hassan; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacology; CHS 828; in vitro; in vivo; hollow fiber; models; NF-kB; Farmakologi; Pharmacological research; Farmakologisk forskning;

    Abstract : The novel cyanoguanidine CHS 828 has shown promising antitumor activity in many in vitro and in vivo studies. The long-term 14 days in vitro hollow fiber cultures, where tumor cells from different tumor cell lines were cultured inside semipermeable fibers, were more resistant to CHS 828 and other cytotoxic drugs than the shorter-term 3 days cultures.
