  1. 1. Protein folding without loops and charges

    Author : Martin Kurnik; Mikael Oliveberg; Kevin W. Plaxco; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; protein folding; folding cooperativity; protein aggregation; protein charges; protein engineering; Biochemistry; biokemi;

    Abstract : Going down the folding funnel, proteins may sample a wide variety of conformations, some being outright detrimental to the organism. Yet, the vast majority of polypeptide molecules avoid such pitfalls. Not only do they reach the native minimum of the energy landscape; they do so via blazingly fast, biased, routes. READ MORE

  2. 2. The birth and growth of the protein folding nucleus : Studies of protein folding focused on critical contacts, topology and ionic interactions

    Author : Linda Hedberg; Mikael Oliveberg; Christopher Johnson; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; protein folding; protein stability; two-state folding; chevron plot; transition-state movements; Hammond behaviour; topology; electrostatic interactions; pH dependence; mass-action; protein engineering; pKa; Biochemistry; Biokemi; biokemi; Biochemistry;

    Abstract : Proteins are among the most complex molecules in the cell and they play a major role in life itself. The complexity is not restricted to just structure and function, but also embraces the protein folding reaction. READ MORE

  3. 3. Folding of the Ribosomal protein S6 : The role of sequence connectivity, overlapping foldons, and parallel pathways

    Author : Ellinor Haglund; Mikael Oliveberg; Neil Ferguson; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; protein folding; protein stability; two-state folding; S6; chevron plot; transition state; parallell pathways; foldon; two-channel landscape; protein engineering; H D-exchange; Biochemistry; Biokemi; Biochemistry; biokemi;

    Abstract : To investigate how protein folding is affected by sequence connectivity five topological variants of the ribosomal protein S6 were constructed through circular permutation.  In these constructs, the chain connectivity (i.e. the order of secondary-structure elements) is changed without changing the native-state topology. 4. 4. The Interaction Between the Chaperonin GroEL and Human Carbonic Anhydrase II : Protein Folding and Stability Studies

    Author : Malin Persson; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; carbonic anhydrase; protein folding; protein stability; chaperone; GroEL; folding mechanism; site-directed spinlabeling; electron paramagnetic resonance EPR ;

    Abstract : The presence of GroEL/ES during the refolding of human carbonic anhydrase II was found to increase the yield of active enzyme from approx. 70 to approx. 100%. This chaperone action on the enzyme could be obtained by adding GroEL alone, and the time course in that case was only moderately slower than the spontaneous process. READ MORE

  5. 5. Structure determination and thermodynamic stabilization of an engineered protein-protein complex

    Author : Elisabet Wahlberg; Per-Åke Nygren; Bengt-Harald Jonsson; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; affibody; protein structure; coupled folding; NMR spectroscopy; protein stability; protein-protein interactions; binding thermodynamics; isothermal titration calorimetry; protein engineering; Structural biochemistry; Strukturbiokemi;

    Abstract : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain.
