1. Structural Adaptation of DNA to Small Ligands and Recombination Proteins
Abstract : In order to contribute to the understanding of mechanisms of recognition in nucleic acid systems, the interaction of DNA with a variety of ligands, representing different cases of adaptation of the double-helix structure, was studied using flow linear dichroism (LD), circular dichroism (CD), fluorescence and absorption spectroscopic techniques. A chiral hydrophobic metal complex, [Ru(dpphen)3]2+ was examined, since this had been reported to be the most efficient molecule to discriminate DNA handedness. READ MORE

3. NMR structral studies on ruthenium complexes binding to DNA
Abstract : The Ru-complex Λ,Λ-[bidppz(bipy)4Ru2]4+ (Λ,Λ-B) exhibits multiple modes of interactions with the palindromic DNA dodecamer [d(CGCGAATTCGCG)]2, however excluding any type of intercalation as shown by both optical spectroscopy and NMR. NMR observation of Λ,Λ-B titration into a DNA solution results in both a transient binding, with observation of fast exchange for protons located near the ends of the DNA duplex, and a more stable interaction in the DNA minor groove indicated by a slow exchange mechanism and binding sites for G2, G4 and A5 of the same DNA strand. 5. DNA Interactions with Chiral Polyaza-aromatic Ruthenium(II) Complexes
Abstract : The binding to DNA of a series of chiral ruthenium complexes is studied with optical spectroscopy techniques, including isotropic absorption, linear dichroism (LD) and emission anisotropy as well as steady-state and time-resolved luminescence. The thesis focuses on the ìlight-switchî complex [Ru(phen)2dppz]2+ which binds to DNA by intercalation and luminesces when bound to DNA but not in pure buffer solution (phen=1,10-phenanthroline; dppz=dipyrido[3,2-a:2',3'-c]phenazine).
