  1. 1. Rational redesign of Candida antarctica lipase B

    Author : Anders Magnusson; Karl Hult; Bernhard Hauer; KTH; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Candida antarctica lipase B; rational redesign; secondary alcohols; substrate-assisted catalysis; S-selective; entropy; aldolase; stereospecificity pocket; oxyanion hole.; Biokemi; Biochemistry; Biokemi;

    Abstract : This thesis describes the use of rational redesign to modify the properties of the enzyme Candida antarctica lipase B. Through carefully selected single-point mutations, we were able to introduce substrate-assisted catalysis and to alter the reaction specificity. READ MORE

  2. 2. Transaminase Biocatalysis: Applications and Fundamental Studies

    Author : Federica Ruggieri; Per Berglund; Mariarita Bertoldi; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; biocatalysis; enzymatic cascade; transaminase; amine; racemization; PLP; enzyme engineering; Chromobacterium violaceum; protein stability; enzyme stability; molecular dynamics; structural biology; enzyme technology; chiral amines; kinetic resolution; Bioteknologi; Biotechnology;

    Abstract : Biocatalysis is the branch of science at the intersection between chemistry and biology and specifcally dedicated to the application of natural evolvable catalysts, i.e. enzymes, in human-designed chemical processes. Among the array of promising biocatalysts, transaminases (EC 2. READ MORE

  3. 3. Heterogeneous catalysis in racemization and kinetic resolution along a journey in protein engineering

    Author : Tamás Görbe; Jan-Erling Bäckvall; Frank Hollmann; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; racemization; protein engineering; kinetic resolution; heterogeneous catalysis; biohybrid catalyst; organisk kemi; Organic Chemistry;

    Abstract : The first part of my thesis concerns the use of heterogeneous acidic resins for racemization of tert-alcohols without any side-product formation. The focus was to develop a system which can be further extended to a DKR protocol consisting of an enzymatic KR reaction. READ MORE

  4. 4. Lipase and ω-Transaminase : Biocatalytic Investigations

    Author : Maria Svedendahl; Per Berglund; Stefan Stefan Lutz; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Candida antarctica lipase B; enzyme catalysis; enzyme catalytic promiscuity; molecular modeling; ω-transaminase; Biochemistry; Biokemi;

    Abstract : In a lipase investigation, Candida antarctica lipase B (CALB) are explored for enzyme catalytic promiscuity. Enzyme catalytic promiscuity is shown by enzymes catalyzing alternative catalytic transformations proceeding via different transition state structures than normal. READ MORE

  5. 5. Transition Metal- and Enzyme-Catalyzed Reactions of Primary Amines and Allylic Alcohols : Powerful Tools in the Preparation of Pharmaceutically Important Compounds

    Author : Lisa K. Thalén; Jan-Erling Bäckvall; Dirk De Vos; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Dynamic kinetic resolution; racemization; catalysis; hydrogen transfer; kinetic resolution; norsertraline; alpha-methyl carboxylic acids; pipecoloxylidide; organisk kemi; Organic Chemistry;

    Abstract : Dynamic kinetic resolution (DKR) has been applied to primary amines utilizing a ruthenium catalyst and an (R)-selective lipase to provide the corresponding amides in up to 95% isolated yield and up to >99% ee. Further investigations of the chemoenzymatic DKR of amines show that it is general and can be applied to a wide range of amines. READ MORE
