  1. 1. Reducing Attrition via Improved Strategies for Pre-clinical Drug Discovery : SPR-biosensor Aided Interaction Studies

    Author : Johan Winquist; Helena Danielson; Roderick E. Hubbard; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biokemi; Biochemistry;

    Abstract : The efficacy of a drug is tightly intertwined with its interaction mechanism with the drug target. The mechanism is dependent on the physicochemical and structural characteristics of both target and drug molecule.Drug discovery is plagued by a high attrition rate, whereas in the clinic, a major issue is drug resistance. 2. 2. Biosensor Studies of Ligand Interactions with Structurally Flexible Enzymes : Applications for Antiviral Drug Development

    Author : Matthis Geitmann; U. Helena Danielson; Marc H. V. van Regenmortel; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; SPR biosensor; HIV-1 reverse transcriptase; HCMV protease; interaction kinetics; drug discovery; non-nucleoside inhibitor; resistance; Biokemi; Biochemistry; Biokemi;

    Abstract : The use of a surface plasmon biosensor fills a missing link in kinetic studies of enzymes, since it measures directly the interaction between biomolecules and allows determination of parameters that are determined only indirectly in activity assays. The present thesis deals with kinetic and dynamic aspects of ligand binding to two viral enzymes: the human cytomegalovirus (HCMV) protease and the human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). 3. 3. Biosensor surface chemistry for oriented protein immobilization and biochip patterning

    Author : Emma Ericsson; Karin Enander; Sofia Svedhem; Linköpings universitet; []
    Keywords : Biochip; biosensor; hydrogel; ligand; orientation; patterning; photochemistry; protein immobilization; surface chemistry; self-assembled monolayer; SAM;

    Abstract : This licentiate thesis is focused on two methods for protein immobilization to biosensor surfaces for future applications in protein microarray formats. The common denominator is a surface chemistry based on a gold substrate with a self-assembled monolayer (SAM) of functionalized alkanethiolates. READ MORE

  4. 4. Phospholipid membranes in biosensor applications : Stability, activity and kinetics of reconstituted proteins and glycolipids in supported membranes

    Author : Inga Gustafson; Gertrud Puu; Sven Lindskog; Peter Konradsson; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Phospholipid films; Liposomes; Membrane proteins; Glycolipids; Ellipsometry; Langmuir Blodgett LB ; Surface plasmon resonance SPR ; Resonant mirror RM ; Atomic force microscopy AFM ; Biosensors; Biokemi; Biochemistry; Biokemi; Biochemistry; biokemi;

    Abstract : In this study the formation of supported membranes onto planar solid supports has been investigated. The stability and activity of reconstituted membrane receptors has been studied. The potential use of such preparations in biosensor applications is discussed.The lipid films were made by the Langmuir Blodgett and by the liposome fusion techniques. READ MORE

  5. 5. Biomolecules at Interfaces -Studied by Surface and Solution Techniques

    Author : Anna Bergstrand; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; LDL; sepsis; PEG; LPS; lipopolysaccharide; QCM-D; SPR;

    Abstract : The interfacial behaviour of biomolecules in general and proteins in particular is interesting from a biochemical point of view since proteins are involved in processes such as complement activation, blood coagulation and atherosclerotic plaque formation, but also in the context of biomedical applications such as solid-phase diagnostics, therapeutic aphaeresis and biosensors. The objective of this thesis is to study biomolecular interactions using surface sensitive biosensor techniques such as surface plasmon resonance, SPR, in combination with bulk techniques. READ MORE
