  1. 1. Structural modeling of membrane transporter proteins

    Author : Jonas Almqvist; Sven Hovmöller; David van der Spoel; Stockholms universitet; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; membrane protein structure; membrane protein crystallography; membrane protein structure modelling; Structural biochemistry; Strukturbiokemi; strukturkemi; Structural Chemistry;

    Abstract : A fundamental process of all living organisms - the transport of molecules across cellular membranes through membrane transport proteins - is investigated.After a brief review of general properties of biological membranes follows a recollection of the major methods of membrane transport that Nature utilizes (Chapter 1). 2. 2. Structural and thermodynamical basis for molecular recognition between engineered binding proteins

    Author : Jakob Dogan; Per-Åke Nygren; John E. Ladbury; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; protein structure; induced fit; binding thermodynamics; NMR spectroscopy; protein engineering; protein-protein interactions; protein stability; calorimetry; Structural biochemistry; Strukturbiokemi;

    Abstract : The structural determination of interacting proteins, both as individual proteins and in their complex, complemented by thermodynamical studies are vital in order to gain in-depth insights of the phenomena leading to the highly selective protein-protein interactions characteristic of numerous life processes. This thesis describes an investigation of the structural and thermodynamical basis for molecular recognition in two different protein-protein complexes, formed between so-called affibody proteins and their respective targets. 3. 3. A structural genomics pilot project : methods and applications

    Author : Ulrika Ericsson; Pär Nordlund; Elisabeth Sauer-Eriksson; Stockholms universitet; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Structural biochemistry; Strukturbiokemi;

    Abstract : With the availability of many completely sequenced genomes, scientific research has shifted from genes to the products of the genes, the proteins. Structural genomics groups have been established worldwide, with the objective of determining protein structures on a genome-wide scale. READ MORE

  4. 4. Protein production and purification in structural genomics

    Author : Martin Hammarström; Per-Åke Nygren; Torleif Härd; Helena Berglund; Herman van Tilbeurgh; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Recombinational cloning; Escherichia coli; gene expression; fusion proteins; solubility; circular dichroism; nuclear magnetic resonance; Structural biochemistry; Strukturbiokemi;

    Abstract : The number of gene products available for structural and functional study is increasing at an unprecedented rate as a result of the successful whole genome sequencing projects. Systematic structure determination of proteins on a genomic scale, called structural genomics, can significantly contribute to the field of protein science and to functional annotation of newly identified genes. 5. 5. Molecular principles of protein stability and protein-protein interactions

    Author : Christofer Lendel; Per-Åke Nygren; Christopher M. Dobson; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; affibody; binding thermodynamics; induced fit; molten globule; NMR spectroscopy; protein engineering; protein-protein interactions; protein stability; protein structure.; Structural biochemistry; Strukturbiokemi;

    Abstract : Proteins with highly specific binding properties constitute the basis for many important applications in biotechnology and medicine. Immunoglobulins have so far been the obvious choice but recent advances in protein engineering have provided several novel constructs that indeed challenge antibodies. READ MORE
