  1. 1. On bacterial formats in protein library technology

    Author : Per-Åke Löfdahl; Per-Åke Nygren; Mikael Widersten; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Affibody molecule; β-lactamase; combinatorial library; green fluorescent protein GFP ; protein engineering; protein fragment complementation assay PCA ; selection; TEVprotease; TNF-α; Bioengineering; Bioteknik; Enzyme engineering; Enzymteknik;

    Abstract : Millions of years of evolution have resulted in an immense number of different proteins, which participate in virtually every process within cells and thus are of utmost importance for allknown forms of life. In addition, there are several examples of natural proteins which have found use in applications outside their natural environment, such as the use of enzymes infood industry and washing powders or the use of antibodies in diagnostic, bioseparation or therapeutic applications. Author : Fredrika Gullfot; Harry Brumer; Tuula Teeri; Jerry Ståhlberg; Mats Martinelle; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; glycosynthase; xyloglucan; xyloglucan endo-transglycosylase; retaining glycoside hydrolase; xyloglucanase; polysaccharide synthesis; Bioengineering; Bioteknik; Enzyme engineering; Enzymteknik; Bioorganical synthesis; Bioorganisk syntes; Biomaterials; Biomaterial;

    Abstract : Xyloglucans are polysaccharides found as storage polymers in seeds and tubers, and as cross-linking glycans in the cell wall of plants. Their structure is complex with intricate branching patterns, which contribute to the physical properties of the polysaccharide including its binding to and interaction with other glycans such as cellulose. 3. 3. On the engineering of proteins: methods and applications for carbohydrate-active enzymes

    Author : Fredrika Gullfot; Harry Brumer; Antoni Planas; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; enzyme engineering; rational design; directed evolution; DNA shuffling; glycosynthase; xyloglucan; xyloglucan endo-transglycosylase; retaining glycoside hydrolase; xyloglucanase; carbohydrate binding module; polysaccharide synthesis; Bioengineering; Bioteknik;

    Abstract : This thesis presents the application of different protein engineering methods on enzymes and non-catalytic proteins that act upon xyloglucans. Xyloglucans are polysaccharides found as storage polymers in seeds and tubers, and as cross-linking glucans in the cell wall of plants. READ MORE

  4. 4. Engineering carbonic anhydrase for highly selective ester hydrolysis

    Author : Gunnar Höst; Bengt-Harald Jonsson; Lars-Göran Mårtensson; Per Berglund; Linköpings universitet; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; carbonic anhydrase; specificity; hydrolysis; rational design; protein engineering; plasmodium falciparum; Bioengineering; Bioteknik;

    Abstract : I denna avhandling presenteras arbete utfört med enzymet humant karboanhydras II (HCAII). Enzymer är en typ av proteiner som accelererar (katalyserar) kemiska reaktioner, vilket är nödvändigt för allt levande. Den naturliga funktionen för HCAII är att katalysera omvandlingen av gasen koldioxid till vätekarbonat, som är löslig i vätska. READ MORE

  5. 5. Enthalpy and Entropy in Enzyme Catalysis : A Study of Lipase Enantioselectivity

    Author : Jenny Ottosson; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biocatalysis; enzyme catalysis; Candida antarctica lipase B; enantioselectivity; enthalpy; entropy; CALB; enantiomeric ratio; Bioengineering; Bioteknik;

    Abstract : Biocatalysis has become a popular technique in organic synthesis due to high activity and selectivity of enzyme catalyzed reactions. Enantioselectivity is a particularly attractive enzyme property, which is utilized for the production of enantiopure substances. READ MORE
