  1. 1. Protein-RNA interplay in translation. Structural studies of RRF, SelB and L1

    Author : Maria Selmer; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Molecular biophysics; Molekylär biofysik; crystallography; mimicry; translation factor; L1; SelB; protein synthesis; RRF;

    Abstract : Translation is a central biological process, that is catalysed by a machinery consisting of RNA and protein. In this thesis three different protein components of the bacterial protein synthesis system have been structurally studied. READ MORE

  2. 2. Protein production, characterization and structure determination in structural genomics

    Author : Esmeralda A. Woestenenk; Torleif Härd; Göran Karlsson; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biology; protein production; protein solubility; fusion tags; nuclear magnetic resonance; structure determination; Biologi; Biology; Biologi;

    Abstract : This thesis covers the process from expression of a heterologous gene in Escherichia coli to structure determination of a protein by nuclear magnetic resonance (NMR) spectroscopy. The first part concerns structural genomics-related parallel screening studies on the effect of fusion tags (in particular the His tag) on protein solubility and the use of fusion tags in fast, parallel purification protocols intended for initial biophysical characterization of human proteins produced in E. 3. 3. Proton transfer in nitric oxide reducing heme-copper oxidases

    Author : Joachim Reimann; Pia Ädelroth; Peter Brzezinski; Michael Wilson; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; heme-copper oxidases; nitric oxide; proton transfer; electron transfer; proton-transfer pathway; Biochemistry; biokemi;

    Abstract : Heme-copper oxidases (HCuOs) are best known as terminal oxidases in the aerobic respiratory chain, in which they catalyze the reduction of oxygen to water. By receiving protons and electrons from opposite sides of the membrane as well as pumping protons, HCuOs contribute to the electrochemical proton gradient over the membrane that can be used for ATP synthesis. 4. 4. Improving initial xylose metabolism in recombinant Saccharomyces cerevisiae

    Author : Márk Gárdonyi; Teknisk mikrobiologi; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biokemisk teknik; Biochemical technology; xylose isomerase; xylose transport; xylose fermentation; Saccharomyces cerevisiae; metabolic engineering;

    Abstract : The aim of the thesis is to improve the initial steps of xylose metabolism in recombinant Saccharomyces cerevisiae. S. cerevisiae takes up xylose of poor affinity by means of hexose transporters. Metabolic control analysis was used to investigate whether the low xylose utilisation rate is due to inefficient transport. READ MORE

  5. 5. Kinetics of proton and electron transfer in heme-copper oxidases

    Author : Peter Lachmann; Pia Ädelroth; Örjan Hansson; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Heme-copper oxidase; electron transfer; proton transfer; nitric oxide reductase; ba3 oxidase; flow-flash; laser-flash photolysis; Biochemistry; biokemi;

    Abstract : Heme-copper oxidases are transmembrane proteins that are found in aerobic and anaerobic respiratory chains. During aerobic respiration, these enzymes reduce dioxygen to water. The energy released in the reaction is used to transport protons across a biological membrane. READ MORE
