  1. 1. The hematopoietic transcription factor RUNX1 : a structural view

    Author : Stefan Bäckström; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Molecular medicine; RUNX1; Runt domain; CBFβ; transcription factor; leukaemia; protein crystallography; anomalous diffraction; Molekylärmedicin; Molecular medicine genetics and pathology ; Molekylär medicin genetik och patologi ; molekylärbiologi; Molecular Biology;

    Abstract : The malfunction of the transcriptional regulator RUNX1 is the major cause of several variants of acute human leukemias and its normal function is to regulate the development of the blood system in concert with other transcriptional co-regulators. RUNX1 belongs to a conserved family of heterodimeric transcription factors that share a conserved DNA binding domain, the Runt domain (RD), named after the first member of this group – Runt - found in Drosophila melanogaster. 2. 2. lac of Time : Transcription Factor Kinetics in Living Cells

    Author : Petter Hammar; Johan Elf; Antoine van Oijen; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; gene regulation; transcription factor; lac operon; facilitated diffusion; single molecule imaging; Molecular Biotechnology; Molekylär bioteknik;

    Abstract : Gene regulation mediated by transcription factors (TFs) is essential for all organisms. The functionality of TFs can largely be described by the fraction of time they occupy their regulatory binding sites on the chromosome. DNA-binding proteins have been shown to find their targets through facilitated diffusion in vitro. READ MORE

  3. 3. Regulation of immunoglobulin transcription

    Author : David Liberg; Immunologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; N-substituted benzamides; NFkB; transcription; B cells; immunoglobulin; Immunology; serology; transplantation; Immunologi; serologi;

    Abstract : Each Ig V-region has its own upstream promoter and the promoter of a mature Ig gene is thereby selected during the V(D)J recombination process. Upon comparison, it is evident that only one transcription factor binding site, the octamer, is preserved within all promoters. READ MORE

  4. 4. Imaging of nucleocytoplasmic transcription factor dynamics in yeast

    Author : Kristofer Bodvard; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; transcription factor; single-cell; yeast; image analysis; Saccharomyces cerevisiae; light stress; Fluorescence microscopy; signalling;

    Abstract : Fluorescence microscopy has proven to be an invaluable tool in identifying and understanding cell signalling, intracellular spatial and temporal dynamics, and cell-to-cell variability. This thesis focuses on imaging of nucleocytoplasmic transcription factor dynamics at the single cell level in yeast. READ MORE

  5. 5. Structure-function relationships of the human Runx1 transcription factor

    Author : Magnus Wolf-Watz; KTH; []
    Keywords : Runx1; Runt domain; CBFâ; transcription; leukemia; protein structure; protein dynamics;

    Abstract : Runx transcription factors contain a DNA-binding andtransactivating α-subunit and a β-subunit (CBFβ)that serves to increase the DNA binding affinity of theα-subunit. The DNA binding domain of Runx proteins isnamed the Runt homology domain (RD) after its strong sequencehomology to theDrosophilaprotein Runt. READ MORE
