    Abstract : Pharmacometric modelling is widely used in many aspects related to type 2 diabetes mellitus (T2DM), for instance in the anti-diabetes drug development, and in quantifying the disease progression of T2DM.The aim of this thesis were to improve the design of early phase anti-diabetes drug development studies with the focus on the power to identify mechanism of drug action (MoA), and to characterize and quantify the progression from prediabetes to overt diabetes, both the natural progression and the progression with diet and exercise interventions, using pharmacometrics modelling. 2. 2. Semi-mechanistic models of glucose homeostasis and disease progression in type 2 diabetes

    Author : Steve Choy; Maria Kjellsson; Mats Karlsson; Willem de Winter; Andrea Mari; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacokinetics; pharmacodynamics; pharmacometrics; glucose homeostasis; insulin; type 2 diabetes; obesity; weight; visceral adipose tissue; HbA1c; non-linear mixed effects; modelling; disease progression; ZDSD rats; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : Type 2 diabetes mellitus (T2DM) is a metabolic disorder characterized by consistently high blood glucose, resulting from a combination of insulin resistance and reduced capacity of β-cells to secret insulin. While the exact causes of T2DM is yet unknown, obesity is known to be a major risk factor as well as co-morbidity for T2DM. READ MORE

  3. 3. [68Ga]Exendin-4: Bench-to-Bedside : PET molecular imaging of the GLP-1 receptor for diabetes and cancer

    Author : Ram kumar Selvaraju; Olof Eriksson; Olle Korsgren; Lars Johansson; Martin Gotthardt; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; PET; [68Ga]Exendin-4; beta cell imaging; insulinoma; dosimetry;

    Abstract : Diabetes epidemic is underway. Beta cell dysfunction (BCF) and loss of beta cell mass (BCM) are known to be key events in its progression. Currently, there are no reliable techniques to estimate or follow the loss of BCM, in vivo. READ MORE

  4. 4. Adherence to lipid-lowering medications and cardiovascular disease prevention in type 2 diabetes mellitus

    Author : Sofia A. Karlsson; Göteborgs universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; medication adherence; refill adherence; medication persistence; pharmacoepidemiology; lipid-lowering medications; type 2 diabetes mellitus; cardiovasular disease; all-cause mortality; cardiovascular mortality; guideline adherence;

    Abstract : Background and aims: Globally, cardiovascular disease (CVD) is the major cause of death among patients with type 2 diabetes mellitus (T2DM). Improved control of LDL cholesterol with lipid-lowering medications and patients’ adherence to such medications have been shown associate with lower risk of CV events and mortality among T2DM patients. 5. 5. Pharmacometric Models of Glucose Homeostasis in Healthy Subjects and Diabetes Patients

    Author : Rikke Meldgaard Røge; Maria Kjellsson; Niels Rode Kristensen; Søren Klim; Mats Karlsson; Roman Hovorka; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Glucose homeostasis; Type 2 diabetes; IGI model; Mechanismbased; NONMEM; pharmacometrics;

    Abstract : Diabetes is a group of metabolic diseases characterized by hyperglycaemia resulting from defects in insulin secretion, insulin action, or both. Several models have been developed for describing the glucose-insulin system. READ MORE
