    Abstract : The thesis work, aimed at the development of laser-spectroscopic detection schemes, consists of three parts, concerned with the development and application of different techniques. The development of UV filtered Rayleigh scattering (FRS) for thermometry and fuel/air ratio measurements, using a frequency tripled, single-longitudinal-mode alexandrite laser at ~254 nm together with an isotopically enhanced mercury filter, is examined first.     Abstract : This thesis exemplifies the strength in the combination of inductively coupled plasma (ICP) spectrometry and electrospray ionization tandem mass spectrometry ESI-MS/MS as detection techniques for liquid chromatography (LC) in the search for and investigation of compounds that bind or can bind hetero-atoms.     Abstract : Cancer is a disease that is strongly related to evolution, as mutations that confer a benefit to individual cells face positive selection and eventually lead to tumorigenesis. As such, the search for genes that drive cancer development entails distinguishing positive selection from other sources of increased mutation rates, which requires detailed knowledge of how normal mutation rates vary across the genome.     Abstract : Two miniaturised atmospheric pressure chemical ionisation (APCI) interfaces were developed for micro column separation and their performances were investigated in high temperature open tubular column liquid chromatography and capillary supercritical fluid chromatography (SFC). The most influential parameters, for example, the interface probe position relative to the sampling orifice and the reagent ion plasma composition, were investigated to optimise the ion current signal and stability.
