1. Exploring molecular interactions between polypeptide conjugates and protein targets : Manipulating affinity by chemical modifications
Abstract : In this thesis molecular interactions between polypeptide conjugates and protein targets were investigated. Polypeptides were derivatized with small organic molecules, peptides and oligonucleotides. New strategies were developed with the aim to increase affinities for proteins of biological interest. 2. Fc receptors and feedback enhancement of antibody responses
Abstract : Antibodies are important for the defence against microorganisms, but are also involved inpathological reactions such as allergy or autoimmunity. The exact mechanisms involved inregulation of antibody responses is still largely unknown. In this thesis, immune regulationby antibodies and the receptors they bind to (FcR), were investigated. 3. Elicitation and enhancement of T and B cell responses
Abstract : The Major histocompatibility complex class I (MHC-I) has been characterized in such great depth that a number of its key properties are well understood and part of its behavior can even be predicted. It is therefore intriguing that the impact of small epitope modifications on immunogenicity and the elicitation of T-cell repertoires remain often unpredictable and full of surprises. READ MORE

4. Feedback Enhancement of Antibody Responses via Complement and Fc Receptors
Abstract : IgG, IgM and IgE in complex with antigen have the capacity to regulate specific immune responses. In this investigation, the role of Fc receptors for IgG (FcγRI, FcγRII and FcγRIII) and complement receptors 1 and 2 (CR1/2) for antibody-mediated enhancement of antibody responses are investigated. 5. Conformationally Constrained Nucleosides, Nucleotides and Oligonucleotides : Design, Synthesis and Properties
Abstract : This thesis is based on six original research publications describing synthesis, structure and physicochemical and biochemical analysis of chemically modified oligonucleotides (ONs) in terms of their potential diagnostic and therapeutic applications. Synthesis of two types of bicyclic conformationally constrained nucleosides, North-East locked 1',2'-azetidine and North locked 2',4'-aza-ENA, is described. READ MORE
