  1. 1. Cytotoxic Cyclotides : Structure, Activity, and Mode of Action

    Author : Erika Svangård; Per Claeson; Lars Bohlin; Ulf Göransson; John H. Cardellina II; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacognosy; cyclotide; cytotoxic; cancer; cyclic cystine knot; Viola; Violaceae; liposome; hollow fibre; homology modelling; mass spectrometry; amphipathic; structure–activity; tumour; membrane disruption; necrosis; Farmakognosi; Pharmacognosy; Farmakognosi;

    Abstract : Cyclotides are small cyclic plant proteins, and this thesis addresses their cytotoxic structure-activity properties and their mode of action on human cancer cell lines. Cyclotides were isolated from Viola odorata and Viola tricolor; three novel cyclotide sequences and two known sequences, but of new origin, were identified using mass spectrometry, amino acid analysis, and Edman degradation. 2. 2. A computational approach to curvature sensing in lipid bilayers

    Author : Federico Elías-Wolff; Alexander Lyubartsev; Max Berkowitz; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; curvature sensing; membrane curvature; cardiolipin; amphipathic helix; symmetric multimers; lipid bilayer; molecular dynamics; Biophysics; biofysik;

    Abstract : Local curvature is a key driving force for spatial organization of cellular membranes, via a phenomenon known as membrane curvature sensing, where the binding energy of membrane associated macromolecules depends on the local membrane shape. However, the microscopic mechanisms of curvature sensing are not well understood. READ MORE

  3. 3. STRUCTURAL, FUNCTIONAL AND EVOLUTIONARY STUDIES OF ANTIMICROBIAL PEPTIDES

    Author : Mukesh Pasupuleti; Lund Dermatologi och venereologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; amphipathicity and end-tagging protease susceptibility; anaphylatoxin C3a; P. aeruginosa; S. aureus; innate immunity; amphipathic molecules; Antimicrobial peptides; infectious disease; bacteriostatic; microbicidal and cytolytic properties;

    Abstract : Antimicrobial peptides represent a heterogeneous group that displays multiple modes of action such as bacteriostatic, microbicidal and cytolytic properties that are sequence and concentration dependent. Life threatening infectious disease is now a worldwide crisis and treating them effectively is becoming difficult day by day, due to the emergence of antibiotic resistant strains at alarming rates. 4. 4. Studies of protein structure, dynamics and protein-ligand interactions using NMR spectroscopy

    Author : Tobias Tengel; Göran Larsson; Maria Sunnerhagen; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; 19F NMR; 2S albumin; PapD; protein-ligand interaction; structure; TFE; Yersinia; YopD; allergy; amphipathic helix; Ber e 1; Brazil nut; dynamics; FimC; LcrH; NMR screening; Biophysics; Biofysik;

    Abstract : In the first part of the thesis, protein-ligand interactions were investigated using the chaperone LcrH, from Yersinia as target protein. The structure of a peptide encompassing the amphipathic domain (residue 278-300) of the protein YopD from Yersinia was determined by NMR in 40% TFE. READ MORE

  5. 5. A Chloroplast Localized Small Heat Shock Protein, Hsp21. Importance of Hsp21 in stress protection of transgenic Arabidopsis thaliana, recombinant expression and characterization of oxidation-dependent conformational changes

    Author : Ulrika Härndahl; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; a-crystallin; sHsp; chaperone; amphipathic a-helix; oligomer; redox-regulated; methionine sulfoxidation; oxidative stress; regulation; protein structure; circular dichroism; mass spectrometry; fluorescence spectroscopy; Biochemistry; Metabolism; Biokemi;

    Abstract : A specific group of heat shock proteins, the so called small heat shock proteins (sHsps), exists in all organisms but are especially abundant in plants. The sHsps prevent aggregation of other proteins (substrate proteins) during transient heat and oxidative stress. READ MORE
